摘 要:目的观察钩吻素子抗外周神经损伤致神经病理性疼痛(NPP)中自发痛的效应。方法在大鼠坐骨神经慢性束缚损伤(CCI)NPP模型上,以术侧自发性抬足时间和足姿势评分为指标,观察单次(7.0,1.4,0.28mg/kg)、连续多次(7.0,0.28mg/kg,每日2次,连续7d)皮下注射给予钩吻素子对NPP中自发痛的效应。结果单次皮下注射给予钩吻素子剂量依赖性减少自发性抬足时间,其中7.0mg/kg组与对照组比较差别有统计学意义(P<0.01);单次皮下注射给予钩吻素子剂量依赖性降低足姿势评分,其中1.4,7.0mg/kg组与对照组比较差别有统计学意义(P<0.05和P<0.01)。连续多次给予钩吻素子减少自发性抬足时间,7.0mg/kg组在术后第3,5,7,9d自发性抬足时间均低于对照组(P<0.01);连续多次给予钩吻素子降低足姿势评分,7.0mg/kg组在术后第3,5,7,9d评分均低于对照组(P<0.05或P<0.01),到术后第9d疗效最强。结论钩吻素子具有抗外周神经损伤致NPP中自发痛的效应。Objective To investigate the nerve injury-induced neuropathic pain. Methods effects of Koumine on spontaneous pain of peripheral Rat chronic constriction injury (CCI) model of neuro- pathic pain was used to observe the effects of Koumine administered subcutaneously one time at dose of 7.0, 1.4 or 0.28 mg/kg, or twice a day for 7 consecutive days at 7.0 or 0.28 mg/kg on spontaneous pain indicated by ipsilateral foot-lifting time and postural index. Results Single dose of Koumine reduced both foot-lifting time and the postural index in a dose-dependent manner, in particular, the difference in the foot-lifting time between 7. 0 mg/kg group and untreated control group was statistically significant (P〈0.01) whereas the significant difference in the postural index was observed in both 1.4 and 7.0 mg/kg group as compared with the control group (P〈0.05 and P〈0.01 respectively). Repeated administration of 7.0 mg/kg Koumine shortened spontaneous foot-lifting time at days 3, 5, 7 and 9 post CCI (P〈0.01). Similarly, Koumine repeated dosing at 7.0 mg/kg also ameliorated the postural index at days 3, 5, 7 and 9 after the injury (P〈0.05 or P〈0.01) with the therapeutic effect being most prominent at day 9 after the CCI operation. Conclusion Koumine has pain-relieving effect on spontaneous pain of peripheral nerve injury-induced neuropathic pain.
