钩吻素子无吗啡样药物依赖性  

No Morphine-like Drug Dependence Potential of Koumine

摘　　要：目的评价钩吻素子产生吗啡样药物依赖性的潜力。方法采用离体豚鼠回肠药物依赖模型、小鼠药物依赖催促戒断模型和自然戒断模型,分析钩吻素子诱导吗啡样身体依赖性的潜力;进而通过小鼠条件性位置偏爱实验评价钩吻素子诱导吗啡样精神依赖性的潜力。结果钩吻素子孵育的离体豚鼠回肠未出现吗啡样纳洛酮催促的戒断反应;小鼠连续给予钩吻素子,未出现吗啡样纳洛酮催促的跳跃、体质量下降和自然戒断的体质量下降等戒断反应;也未见吗啡样条件性位置偏爱效应。结论钩吻素子可能无吗啡样药物身体依赖性和精神依赖性的潜力。Objective To evaluate whether Koumine produces morphine-like drug dependence. Methods Morphine-like physical dependence potential of Koumine was evaluated by the drug dependence model of isolated guinea-pig ileum, mouse drug dependence precipitated withdrawal model and natural withdrawal model. Morphine-like psychological dependence potential of Koumine was assessed by the conditioned place preference testing. Results Isolated guinea-pig ileum incubated with Koumine showed no signs of naloxone precipitated morphine-like withdrawal symptoms. Mice repeatedly administered with Koumine showed neither morphine-like naloxone precipitated withdrawal symptoms such as jumping, body weight loss and natural withdrawal body weight loss, nor morphine-like conditioned place preference. Conclusion Koumine might not induce morphine-like physical and psychological dependence.
