摘  要:目的观察氯化镧(LaCl3)对仔鼠学习记忆、海马环磷酸腺苷(cAMP)含量及蛋白激酶A(PKA)表达的影响。方法将60只Wistar雌性大鼠,随机分为对照组(饮用蒸馏水)和低、中、高剂量染镧组,LaCl3染毒组从雌鼠受孕第0天起通过自由饮水染毒,至仔鼠断乳后1个月,然后对仔鼠进行水迷宫实验,并测定仔鼠海马cAMP含量、PKA mRNA和蛋白的表达水平。结果各染镧组仔鼠逃避潜伏期显著高于对照组(P<0.05),随着染毒剂量增加,大鼠游泳距离增加(P<0.05),穿过目标平台次数减少(P<0.05),海马cAMP含量显著降低(P<0.05),PKA mRNA和蛋白表达显著下调(P<0.05)。结论镧损害大鼠学习记忆能力,其机制可能与其所致的大鼠海马组织cAMP含量减少及PKA mRNA、蛋白表达水平下降有关。Objective To study the effects of lanthanum (La) on hippocampal cAMP content and PKA expression in rats, and discuss the mechanism underlying learning and memory impairments induced by La. Methods Sixty female Wistar rats were divided into control (drank distilled water), low-, moderate-, and high-dose LaC13 groups (drank 0. 25%, 0. 5% and 1% LaC13 distilled water, respectively). The pregnant rats were given LaC13 by drinking water from day 0 of pregnancy to one month after weaning of offspring. Morris water maze was used to evaluate the ability of learning and memory of offspring rats. ELISA was used to analyze the cAMP content, and RT- PCR and Western blot were applied to determine the expression of PKA mRNA and protein in the hippocampus of offspring rats respectively. Results In the water maze tests,learning and memory of three La groups were significantly weaker than those of control group. The contents of cAMP, PKA mRNA and protein expression in the hippoeampus of three La groups were signifieantly lower than those of control groups. Conclusion The learning and memory impairments caused by La may be associated with lower content of cAMP and down-regulation of PKA mRNA and protein expression in the hippoeampus.
