摘  要:[目的]探讨在CV-1细胞中肝细胞核因子3(HNF3)对Ⅱ型葡萄糖载体(GLUT2)表达的影响.[方法]利用DNA克隆技术将HNF3cDNA序列重组到表达载体pSV-sport上,GLUT2cDNA克隆到载体pGL3b上.通过CV-1细胞株的体外培养将HNF3转染CV-1培养细胞,应用荧光素酶活性测定和Western blot方法观察HNF3对GLUT2的启动子活性及蛋白质表达的影响.[结果]HNF3可增高GLUT2的荧光素酶活性及蛋白质表达水平.[结论]HNF3可能通过调节GLUT2的表达参与胰岛素对血糖的调节过程.OBJECTIVE To study the effects of hepatocyte nuclear factor 3 (HNF3) on expression of glucose transporter II(GLUT2) in CV-1. METHODS HNF3 cDNA sequence was cloned into pSV-sport expression vector, and GLUT2 cDNA sequence was cloned into pGL3b vector. The recombinant HNF-3 and /or GLUT2 were transfected into CV-1 cells, and the effects of HNF3 on protein expression and promoter activity of GLUT2 were observe by luciferase assay and Western blot. in CV-1 cells confirm the GLUT2 promoter activity with luciferase assay and protein expression with Western blot by time dependent and dose dependent treatment with HNF-3 transfect ion. RESULTS HNF3 increased the GLUT2 promoter activity and protein expression. CONCLUSION HNF3 may be participates in regulating process of insulin for blood glucose level by through regulating GLUT2 expression.
