不同标准品对微生物法检测血浆叶酸结果的影响

摘  要:目的比较采用叶酸和5-甲基四氢叶酸作为标准品对微生物法检测血浆叶酸结果的影响。方法分别采用叶酸和5-甲基四氢叶酸作为标准品并绘制相应的标准曲线,用微生物法测定标准参考物质的高、中、低三个浓度的血浆叶酸水平,比较不同标准曲线对标准参考物质测定结果的影响。结果以5-甲基四氢叶酸标准品做标准曲线,标准参考物质高、中、低浓度测定值与真值的相对误差依次为2.87%、3.60%和-1.92%;以叶酸标准品做标准曲线,相对误差依次为6.77%、8.38%和7.28%。结论以叶酸或5-甲基四氢叶酸作为标准品测定标准参考物质的高、中、低浓度的测定值与真值的相对误差均可接受(<10%),但以5-甲基四氢叶酸为标准品测定标准参考物质的高、中、低浓度的测定值与真值的相对误差较小,测定结果更精准,建议微生物法检测血浆叶酸采用5-甲基四氢叶酸为标准品。Objective To compare the effect of 5 -methyltetrahydrofolate versus folic acid as the calibrator on microbiological assay in plasma folate measurement. Methods Standard procedures of microbiological assay for plasma folate were carried out with folic acid and 5 - methyltetrahydrofolate as calibrator. Three levels (low, medium and high) of standard reference material SRM 1955 were involved in the assay. Absolute and relative differences in plasma folate measurements were calculated and compared between the two calibrators. Results The relative errors between the true value and the measured value at the high, medium and low concentration of standard reference material were 6.77% , 8.38% and 7.28% , respectively, when using folic acid as calibrator, while they were 2.87%, 3.60% and - 1.92%, respectively, when using 5 - methylterahydrofolate as calibrator. Conclusion Although differences in plasma folate concentration were both acceptable ( 〈 10% ) when using folic acid and 5 - methyhetrahydrofolate as a calibrator, plasma folate using 5 - methyhetrahydrofolate as calibrator has higher precision. Therefore, 5 - methyltetrahydrofolate is recommended as the calibrator in plasma folate detection by microbiological assay.
