摘  要:目的观察氧化苦参碱(OMT)对兔血管平滑肌细胞(VSMC)增殖的影响。方法采用高脂诱导兔动脉粥样硬化(AS)模型,体外培养兔主动脉VSMC,以3H-TdR掺入、流式细胞分析和cDNA探针原位杂交方法观察OMT对VSMC增殖和c-fos、c-myc基因表达的影响。结果 OMT可显著抑制VSMC增殖,使处于G1期与G2+M的细胞显著增多,同时抑制VSMC中c-fos和c-myc基因的表达。结论 OMT对VSMC增殖有显著抑制作用,其机制可能与抑制VSMC中c-fos和c-myc基因表达有关。Objective To explore the effect of oxymatrine (OMT) on vascular smooth muscle cells (VSMC) proliferation of' rabbit. Methods A atherosclerosis model of rabbit VSMC induced by high fat was established. The effect of OMT on VSMC proliferation, c -los and c -mye gene expression in the VSMC was observed by 3H -TdR insertion, flow cytometry analysis, cDNA probe - in situ hybridization. Results The VSMC proliferation was depressed by OMT markedly and the c - los, c - myc gene expression can be inhibited by OMT significantly, while the cells of G1 phase and G2 + M were increased. Conclusion The VSMC proliferation can be inhibited by OMT significantly, and the inhibition of the e - fos and c - myc gene expression may contribute to explain the mechanism of OMT inhibition on VSMC proliferation.
