白香丹胶囊对焦虑大鼠模型抗焦虑作用的研究

摘　　要：目的:建立焦虑大鼠模型,探讨白香丹胶囊的抗焦虑作用。方法:采用不确定性空瓶刺激法建立焦虑大鼠模型。应激最后3 d对所有大鼠进行行为学观察,应激结束后以旷场实验和高架十字迷宫实验对其行为学进行评价,通过ELISA实验检测大鼠血清去甲肾上腺素(NA)、脑源性神经营养因子(BDNF)水平。结果:与空白组比较,模型组大鼠处于易激惹状态,表现出频繁的攻击和修饰行为,旷场实验总路径增高(P<0.01),高架十字迷宫实验进入开放臂次数百分数(OE%)和开放臂停留时间百分数(OT%)均降低(P<0.01),ELISA实验所测大鼠血清NA、BDNF水平均增高(P<0.01)。与模型组比较,白香丹胶囊给药组和地西泮给药组大鼠的修饰和攻击行为明显减少,旷场实验总路径降低(P<0.01),OE%和OT%值升高(P<0.05),NA和BDNF水平降低(P<0.01)。结论:不确定性空瓶刺激作为一种慢性心理应激模型,较好模拟了焦虑症的发病状态,白香丹胶囊具有明显的抗焦虑作用。Objective：To establish anxiety model of rats and investigate the anti-anxiety effect of Baixiangdan capsule. Methods：The anxiety model of rats was established by using the stimulation of uncertainty empty bottle. All rats＇ behaviors were observed in the last 3 days of stress. The ethology of rats reviewed by ap- plying international open field test and elevated plus-maze test after the stress. The levels of Noradrenaline （NA）,brain-derived neurotrophic factor（BDNF） in serum were detected by ELISA experiments. Results：Com- pared with the normal group,the model group were irritable and had frequency grooming and attack behav- iors,the total path of open field test increased（P〈0.01）,and the open arm entry percent（OE%） and the open arm time percent（OT% ） of elevated plus-maze test decreased （P〈 0.01） , and the levels of NA and BD- NF were also increased（P〈0.01）. Compared with the model group,the rats＇ grooming and attack behaviors of Baixiangdan capsule group and diazepam group obviously decreased,and the total path of open field test decreased,OE% and OT% increased,and the levels of NA and BDNF decreased. Conclusion：The model of chronic psychological stress by the stimulation of uncertainty empty bottle well simulateed the anxiety symp- toms in rats.Baixiangdan capsule has significant anti-anxiety effect.
