摘 要:目的考察酚红在大鼠不同肠段的吸收特性,及葛根素、维拉帕米对酚红肠吸收的影响,探讨酚红法不适用于研究葛根素肠吸收的机制。方法采用在体单向肠灌流法,HPLC法测定灌流前后酚红的含量,研究分别加入葛根素和维拉帕米后,酚红在各肠段的吸收情况。结果加入维拉帕米后,酚红的吸收有所增加,维拉帕米质量浓度为100μg·mL-1时,与对照组比较,各肠段差异均有统计学意义(P<0.01);加入葛根素后,酚红的吸收有所增加,尤其在结肠中最为明显,差异有统计学意义(P<0.05)。结论酚红在肠道有吸收,且为P-糖蛋白的底物;酚红法不适用于研究葛根素肠吸收机制的原因之一是葛根素能促进酚红在肠道的吸收。Objective To study the absorption characteristics of phenol red in different intestinal sections and the impact of puerarin and verapamil on the phenol red absorption in order to prove phenol red assay being not applicable to the mechanism study of intestinal absorption of puerarin. Methods The rat single pass perfusion technique was applied. The concentration of phenol red was determined by HPLC. The effect of puerarin and verapamil on the absorption of phenol red was studied. Results Phenol red was absorbed rapidly when coadministered with verapamil. There was obvious difference between the test group and the control group when 100 ~g ~ mL 1 verapamil was added ( P 〈 0.01 ). Phenol red was absorbed rapidly when coadministered with puerarin. The best absorption enhancing site of phenol red was colon and there was significant difference between the test group and the control group (P 〈 0.05 ). Conclusions Phenol red is P-gp substrate and can be absorbed. Phenol red assay is not applicable to the mechanism study of intestinal absorption of puerarin, because puerarin can increase the absorption of phenol red.
