摘　　要：源自数种光滑型菌株及 R_a,R_c,R_e 粗糙型菌株的纯化脂多糖水溶液、游离类脂 A 以及多种经煮沸处理的肠道杆菌上清液,在通过 DEz-sepharose 层析柱后鲎试验均转阴性,说明该层析柱可同时吸附多种粗制的或精制的内毒素混和物,其结合位点在类脂 A,吸附容量大于0.8mg/ml 凝胶,且可经脱氧胆酸钠洗脱再生而反复使用。DEz-sepharose 亲和层析为一种去热原质的简便有效的新方法。The removal of different forms of bacterial endotoxins from solutions by DEZ-sepharose affinity chromatography was studied.Solutions of purified lipopolysaccharides from several S-form and Ra,Rc and Re strains,lipid A solution and the supernatants containing endotoxins from five strains of heat- killed enteric bacilli were passed through a DEZ-sepharose column consecutively.No endotoxing was found in the collected effluents as tested by limulus lysate assay.DEZ-sepharose gel was able to bind a variety of endotoxins at the same time.The adsorption capacity was above 0.8mg LPS per ml gel.The bound endotoxins were absorbed from the gel with 1% sodium deoxycholic acid solution;the column was thus used repeatedly.Our results indicate that DEZ-sepharose gel may be capable of removing any forms of endotoxins owing to its affinity with lipid A.As a simple but effective new technique for pyrogen removal,DEZ-sepharose affinity chromatography is of great value for application.
