摘 要:目的观察转化生长因子(TGF)-β1诱导肾小管上皮细胞-间充质细胞转分化(epithelialmesenchymal transition,EMT)时microRNA(miRNA)表达谱的变化及丹酚酸B的干预效应。方法体外培养人近端肾小管上皮细胞系细胞(HK-2),分为3组:(1)对照组:细胞培养基中未加入丹酚酸B或者TGF-β1;(2)TGF-β1组:细胞培养基中加入TGF-β1;(3)丹酚酸B干预组:细胞培养基中分别加入丹酚酸B(50μmol/L)和TGF-β1(浓度为5 ng/ml)。采用基因芯片和RT-PCR检测各组细胞miRNA的表达变化。结果 (1)与正常HK-2细胞比较,TGF-β1组有118种miRNA的表达出现显著改变,其中28种表达下调2倍以上,90种表达上调2倍以上。(2)与TGF-β1组比较,丹酚酸B干预组有256种miRNA的表达出现显著改变,其中172种表达下调2倍以上,84种表达上调2倍以上。(3)有19种miRNA表达在两组同时出现显著改变,其中8种在TGF-β1组明显下调,而在丹酚酸B干预后明显上调,11种在TGF-β1组明显上调,而在丹酚酸B干预后则明显下调。结论肾小管EMT时伴有miRNAs表达谱的明显变化,部分miRNA的表达变化与丹酚酸B的干预密切相关,可能成为治疗肾间质纤维化新的靶点。Objective To investigate the alteration ofmieroRNA (miRNA) expression patterns during the preventive effects of salvianolic acid B (Sal B) on epithelial-mesenchymal transition (EMT) induced by transforming growth faetor-β1 (TGF-β1). Methods In this study, we used Human kidney proximal tubular cell line (HK-2) cultured in vitro as the proximal tubular cell model. Cells were divided into three groups as follows: Control group, TGF-β1 group, Sal B preventive group, miRNAs were detected by gene chips and RT-PCR respectively. Results (1)Compared with normal HK-2 cells, 118 miRNAs expression profiles were significantly changed in TGF-βI group, of which 28 were downregulated and 90 upregulated more than 2 times. (2)Compared with TGF-131 group, 256 miRNAs were showed differentially expressed profiles in Sal B preventive group with 172 downregulated and 84 upregulated more than 2 times. (3)Among the 19 miRNAs altered in both the two groups, 8 were markedly downregulated in TGF-β1 group while upregulated in Sal B preventive group, and 11 dramatically upregulated in TGF-β1 group while dowaregulated in Sal B group. Conclusion EMT of renal tubular prevent by Sal B attaches to different expression of miRNAs, and parts of the miRNAs may be closely associated with the preventive effects.
