摘  要:以"黔椒四号"辣椒带柄子叶为外植体,采用正交实验设计研究了辣椒离体培养中带柄子叶外植体叶状体的发生以及进一步获得伸长芽进而获得再生植株的方法。结果表明:BA、IAA和AgNO3均显著影响带柄子叶的分化,其影响作用大小依次为:BA>AgNO3>IAA,最佳分化培养基为MS+BA 6.0mg/L+IAA 1.0mg/L+AgNO36.0mg/L+蔗糖30g/L+琼脂7g/L,分化率为82%;而叶状体在MS/MB+BA 6.0 mg/L+IAA 0.5 mg/L+GA3(0、1、2、3mg/L)+蔗糖30g/L+琼脂7g/L培养基培养4周可得到伸长的芽;GA3是获得伸长芽不可缺少的添加物质,其浓度(1、2、3mg/L)显著影响平均伸长芽数,对伸长率的影响达到了极显著水平;MB和MS基础培养基对平均伸长芽数和伸长率无显著影响,但MB基础培养基有利于获得更多健壮的伸长芽。MB+BA 6.0mg/L+IAA 0.5mg/L+GA32mg/L+蔗糖30g/L+琼脂7g/L最适宜获得伸长芽,伸长率为36%,获得的伸长芽容易生根形成再生植株。Cotyledons with petioles of 'Qianjiao No. 4' pepper (Capsicum annuum) were used as explants which differentiated into rosettes. The influences of BA, IAA and AgNO3 on cotyledon explants differentiation were tested according orthogonal experiment design. The results showed that the influences of BA, IAA and AgNOa on explants differentiation were significant at 5% level and ranked as BA〉AgNOa 〉IAA. MS+BA 6. 0 mg/L+IAA 1.0 mg/L+ AgNO3 6.0 mg/L+sugar 30 g/L+agar 7 g/L was chosen as the best medium for cotyledon explants differentiation with differentiation rate as 82 %. After four weeks culture on MS/MB+BA 6. 0 mg/L+ IAA 0. 5 mg/L+GAa (0,1,2,3 mg/L) + sugar 30 g/L+agar 7 g/L,the elongated shoots from rosettes were obtained on elongation media. GAa was indispensable for obtainment of elongated shoots, and its concentration (1,2,3 mg/I.) affected the elongation rates at 1% level and affected the average number of elongated shoots at 5% level. The MB basal medium was not better than MS basal medium at the elongation rate and the average number of elongated shoots. But the MB basal medium gave more well- grown shoots than the MS basal medium. MB+BA 6. 0 mg/L+IAA 0. 5 mg/L_GAa 2 mg/L+sugar 30 g/L+agar 7 g/L was chosen as the best elongation medium with elongation rate as 36%. On rooting medium,the elongated shoots rooted well and developed into plants.
