摘  要:CBF基因是植物CBF抗冷途径的枢纽,对增强植物抗冷能力极为重要。本研究根据已知的核桃CBF基因序列设计通用性引物,对麻核桃cDNA进行PCR扩增,成功获得麻核桃CBF基因全长序列,命名为JhCBF。JhCBF全长820 bp,包含一个645 bp的开放读码框(ORF),编码214个氨基酸。序列分析表明JhCBF与其他物种来源的CBF具有较高的同源性,其中,与桦木科白桦的同源性最高,为61.22%;JhCBF含有一个预测的AP2结构域及CBF特有的两段短肽序列。系统进化树分析表明JhCBF与桦木科白桦的亲缘关系最近,最先聚为一类。CBF gene plays a central role in CBF cold-response pathway in plant. It is very important in increasing plant cold tolerance. Universal primers were designed according to published walnut CBF genes,and then PCR amplification was carried out using Juglans hopeiensis cDNA. Finally,the Juglans hopeiensis CBF gene was obtained,named JhCBF. The full-length of JhCBF was 820 bp,containing an opening reading frame( ORF) of 645 bp. The deduced protein had 214 amino acids. Sequence analysis showed that JhCBF shared high homology with CBF from other plants. Among them,the homology with CBF from Betula platyphylla( Betulaceae) was the highest of 61. 22%. JhCBF contained a predicted AP2 domain and two peptide sequences which were specific to CBF. Phylogenetic tree analysis indicated that JhCBF had the closest relationship with CBF from Betula platyphylla( Betulaceae),and they were clustered into one group firstly.
