摘  要:利用PCR和测序技术获得江西安义瓦灰鸡和东乡绿壳蛋鸡MHC B-G基因全序列,分析其序列多态性,并预测其蛋白质结构与功能。结果表明:瓦灰鸡和东乡绿壳蛋鸡MHC B-G基因全序列大小分别为6 427和6 107bp,2个鸡品种存在216个变异位点,均为单一多态位点,共缺失624bp。核苷酸多样度(Pi)为0.037 2,平均核苷酸差异(K)为216。2个鸡品种MHC B-G蛋白的二级结构以无规则卷曲和延伸链为主,且具有明显的跨膜螺旋结构,疏水性、柔韧性、表面可能性及抗原性预测结果显示,该蛋白具有丰富的B细胞抗原位点,分别含有108和85个磷酸化位点。MHC B-G蛋白在2个鸡品种中功能类似,但存在一定差异,2个鸡品种MHC B-G蛋白均在运输与合成功能方面可能性均最大。此外,瓦灰鸡MHC B-G蛋白主要承担生物合成辅因子的功能,在转录方面发挥重要作用;而MHC B-G蛋白在东乡绿壳蛋鸡中主要承担阳离子通道的作用。The whole MHCB-G sequence of Wahui and Dongxiang blue was obtained using PCR and sequencing and its structure and function of the protein was also predicted. The whole sequence of Wahui and Dongxiang blue was 6 427 and 6 107 bp, respectively. There were 216 polymophic sites, all of which were single variable sites. Sites with alignment gaps or missing data were 624 bp in two chicken breeds. Nucleotide diversity (Pi) was 0. 037 2. Average number of nucleotide differences (K) was 216. MHC B-G protein secondary structure had more extended strand and random coil region, with signal transmembrane helix. MHC B-G protein contained 108 and 85 phosphorylation sites in Wahui and Dongxiang blue, respectively. The hydrophobicity structure prediction of the two chicken breeds MHC B-G protein indi- cated that it had major hydrophobicity region. There were more overlapping region in flexibility, surfaceprobability and antigencity, which made antigen and antibody combine easily. The MHC B-G protein function prediction indicated that it had transportation composition in two chicken breeds in maximum probability. In addition, MHC B-G protein mainly takes part in biosynthesis cofacor and transcription in Wahui and mainly has a role in cation gallary in Dongxiang Blue.
