摘 要:为探讨雄性大鼠去睾后骨丢失的组织学机制 ,以及应用益肾健脾胶囊和钙剂后的组织学反应。选用 10月龄大鼠 6 0只 ,行去睾术制成病理模型。随机分为正常组、病理组、益肾健脾胶囊组、益肾健脾 +钙剂组、益肾 +钙剂组和钙剂组。用药 12周后处死取L1~ 2 作不脱钙骨切片 ,测定骨形态计量学各骨量参数和表面参数。各组参数均作方差分析 ,结果①病理组骨量参数小于正常组 (P <0 .0 5 ) ;表面参数显示成骨参数减少 ,破骨参数增加 ,骨转换加快 (P <0 .0 5 )。②各中药组骨量参数高于病理组 (P <0 .0 5 ) ;表面参数得到逆转。同时可见部分成骨细胞特别活跃 ,其下类骨质高出周围静止表面。③益肾健脾 +钙剂组皮质骨厚度和骨内膜矿化率高于其他中药组及钙剂组 (P <0 .0 5 )。表明重建负平衡和骨高转换是导致雄性大鼠去睾后骨丢失的组织学机制。益肾、健脾、补钙同时应用具有抑制骨吸收 ,促进骨形成 ,降低骨转换 ,提高机体对钙剂的吸收利用度 ,从而影响骨矿化过程 。TCM Hospital of Hangzhou City,310007 Objective:In order to explore the histological mechanism of bone loss in the orchiectomized rats and histological reaction after“Yishen Jianpi”capsule and calcium agent are given.Method:The rats aged 10 months were selected to apply orchiectomy and made pathological model,and then randomly divided into 6 groups:Group A as the normal,Group B as the pathological ,Group C treated wity “Yishen Jianpi”capsule, Group D treated with“Yishen Jianpi”capsule matching calcium agent,Group E treated wity kidney-tonified TCM matching calcium agent and Group F treated with calcium agent.The rats were given medicines for 12 weeks,and then killed to take out L 1 and L 2 vertebrae and make no-decalcifyingbone section.The parameter of bone quantity and bone surface in the histomorphometry were determined and made analsis of variance respectively.Result:(1) The parameter of bone quantity in Group B was lower than that of Group A (P< 0.05),it was showed that the parameter of osteogenesis was decreased,osteoclasia parameter was increased and the bone transformation was accelerated (P< 0.05).(2)The parameter of bone quantity in Group C,D and E was higher than that of Group B (P< 0.05);The parameter of surface were reversed, at the same time it was seen that some osteoblasts were quity active, the osteoid under osteoblasts were higher than surrounding static surface.(3) The thickness of cortex of bone and the mineralization rate endosteum in Group D were higher than those of Group C,E and F.Conclusion:Rebuliding negative balance and high bone transformation were histological mechanism of bone loss in the orchiectomized rats.Tonifying the kidney,strengthening the spleen and supplying calcium simultaneously can inhibit the bone absorption,promote the bone formation,dectease the bone transformation,raise the degreeof absorbing and utilizing calcium agent and affect the course of bone mineralization.So it was a best way to treat osteoporosis of the old male.
