摘 要:目的探讨伤科接骨片促兔下颌骨缺损骨组织修复的可能作用机制。方法健康雄性新西兰兔72只,随机分为正常对照组(24只)、模型对照组(24只)和伤科接骨片组(24只),除正常对照组外其余各组建立下颌骨缺损模型。正常对照组、模型对照组给予正常饲料40g/d,伤科接骨片组给予伤科接骨片饲料40g/d(约含伤科接骨片0.528g)。分别于造模后第7、14、28、56天各处死6只兔,行下颌骨侧位X线片检查后,收集下颌骨缺损处骨组织块,检测环氧合酶-2(COX-2)蛋白、COX-2mRNA表达。结果与正常对照组比较,模型对照组造模后第14、28天兔下颌骨组织COX-2蛋白及COX-2mRNA表达明显升高(P<0.05);与模型对照组比较,伤科接骨片组造模后第14、28、56天兔下颌骨组织COX-2蛋白及COX-2mRNA表达明显升高(P<0.05)。影像学显示,伤科接骨片组较模型对照组兔下颌骨缺损区成骨速度更快,新生骨密度更大。结论伤科接骨片可能通过上调COX-2蛋白及COX-2mRNA表达促进兔下颌骨缺损骨组织的修复。Objective To study the possible repair mechanism of Shangke Jiegu Tablets(bone-setting tablets)for mandibular defect.Methods Totally 72healthy male New Zealand rabbits were randomized into the normal control group,model control group and Shangke Jiegu Tablets group,with 24in each.The mandibular defect models were established in all groups except the normal control group.The normal control group and model control group were given 40g/d normal forage.The Shangke Jiegu Tablets group was given 40g/d Shangke Jiegu Tablets forage(containing about 0.528g Shangke Jiegu Tablets).Six rabbits in each group were sacrificed on the 7th,14th,28th and 56th day after modeling respectively.The bone tissue in mandibular defect area was collected after mandible X-ray examination.The protein and mRNA expression of Cyclooxygenase-2(COX-2)and the positive dyeing strength and area of bone tissue were detected.Results Comparing with the normal control group,the protein and mRNA expression of COX-2increased significantly on the 14th and 28th day after modeling in the model control group(P〈0.05).Comparing with the model control group,the protein and mRNA expression of COX-2increased significantly on the 14th,28th and 56th day after modeling in the Shangke Jiegu Tablets group(P〈0.05).Imageology showed that the speed of bone-formation was faster and the density of new bone was greater in mandibular defect area in the Shangke Jiegu Tablets group.Conclusion Shangke Jiegu Tablets can repair the mandibular defect by up-regulating the protein and mRNA expression of COX-2.
