摘 要:目的建立人血浆中拉莫三嗪的HPLC测定方法。方法采用利奈唑胺为内标,用10%高氯酸甲醇溶液沉淀蛋白,色谱柱为Xbrige C18(4.6 mm×150 mm,5μm);流动相为乙腈-5 mmol/L磷酸二氢钾溶液(19∶81);流速为1.0 ml/min,柱温35℃;检测波长:225 nm。结果拉莫三嗪在0.2 mg/L^40 mg/L范围内线性关系良好,r=0.9996,定量下限为0.2 mg/L,平均绝对回收率92.17%,方法回收率在98%~106%之间。高、中、低3种浓度的日内、日间RSD均小于5%。结论本方法简单、快捷、灵敏、准确,适用于拉莫三嗪临床血药浓度的监测。Objective To establish a HPLC method for the detection of lamotrigine in human plasma.Methods Thetarget analytes in plasma were extracted by protein precipitation using 10% perchloric acid methanol,and linezolid was used as the internal standard.The separation was performed on a Xbrige C 18 column(4.6 mm* 150 mm,5 μm) at 35℃.The mobile phase consisted of acetonitrile-5 mmol / L monopotassium phosphate(19∶ 81) with a flow rate of 1.0 ml / min.The UV detection wavelength was 225 nm.Results The calibration curves of lamotrigine showed good linear regression in the range of 0.2 mg / L to 40 mg / L(r = 0.9996).The lower limit of quantification of lamotrigine was 0.2 mg / L.The mean absolute recovery was 92.17%,and the method recovery was 98% ~ 106%.Intra-and inter-day variations were less than 5%.Conclusion The established method is simple,sensitive and accurate for determining Lamotrigine level in human plasma.
