摘 要:目的了解多重PCR、荧光定量PCR及培养分离法检测腹泻患者空肠弯曲菌的差别及增菌过程对检测结果的影响。方法比较多重PCR、荧光定量PCR及培养分离法对192例腹泻患者粪便标本中空肠弯曲菌的检测率与增菌前后的变化。结果荧光定量PCR的检出率最高(增菌前后分别为5.21%、4.17%),多重PCR次之(增菌前后分别为4.69%、3.65%),培养分离法最低(增菌前后分别为1.04%、3.13%)。结论荧光定量PCR、多重PCR比较适合于空肠弯曲菌的监测检测,多重PCR更适合基层单位应用,培养分离方法适合临床治疗患者使用。增菌对不同方法的检出率有一定的影响,应根据不同的检测目的加以选用。Objective To understand the differences of detecting Campylobacter jejuni by multiplex PCR,real time PCR,and traditional culture-based method in diarrhea patients and the influence of enrichment culture on the isolation of C.jejuni in diarrhea patients.Methods Multiplex PCR and real time PCR were applied,along with traditional culture-based method,to detect C.jejuni in 192 diarrhea patients.In parallel,the three detection methods were performed both before and after enrichment.Results The highest percentage was obtained by real time PCR.By real time PCR analysis on the samples before and after enrichment,the positive rate of C.jejuni were 5.21% and 4.17%,respectively.The results obtained by multiplex PCR and traditional culture-based method before and after enrichment were 4.69%,3.65% and 1.04%,3.13%.Conclusion Real time PCR and multiplex PCR were more suitable for the monitoring of C.jejuni.,especially in institutions in rural areas; multiplex PCR was the best choice comparing to Real time PCR.For the clinical treatment of patients,it should give priority to the traditional culture-based method.The influence of enrichment culture on the three detection approaches was different.So the enrichment culture should be used according to our detection purpose.
