摘 要:目的评估这几种检验方法在检验食品中EHEC O157:H7的特异性、敏感性。方法用平板分离培养法、酶联免疫法(ELISA)、胶体金法、荧光PCR法同时对肠出血性大肠杆菌O157:H7进行检测。结果平板分离培养法和荧光PCR法的特异性最好,高浓度时三种检测方法的敏感性接近100%,ELISA法敏感性最低86%。结论显色平板分离培养法具有最好的检测特异性和灵敏度,但检测时间较长;ELISA法灵敏度低,但快速,费用不高,无需仪器,适合基层实验室;胶体金法检测灵敏度高,有假阳性,费用高;荧光PCR法检测敏感性与特异性均较高。Objective To evaluate the specificity and sensitivity of the four methods in detection of EHEC O157 :H7 in food. Methods The enterohemorrhagic escherichia coli O157 : H7 was detected in food by plate culture method, enzyme - linked immunosorbent assay (ELISA), colloidal gold method and fluorescent PCR method. Results Tablet separation and culture method and the fluorescent PCR had best specificity. At high concentrations, ELISA had the lowest sensitivity (86%) , while that of the other three detection methods was close to 100%. Conclusion Color cultutre method has the best detection specificity and sensitivity, but time -consuming; ELISA method has low sensitivity, but rapid, low -cost, without instrument, suitable for primary laboratory; Colloidal gold assay was high sensitive and high - cost with false - positive; The fluorescent PCR assay has higher sensitivity and specificity.
