摘 要:目的研究白黎芦醇对谷氨酸神经毒性损伤的离体大鼠海马CA1区神经元的保护作用及其机制。方法将1.5 mmol·L-1的谷氨酸加入到原代培养的新生大鼠海马CA1区神经元培养液中,制造神经元毒性损伤模型;将20μmol·L-1的白黎芦醇分别加入到原代培养正常的和兴奋性毒性损伤的海马CA1区神经元培养液中,观察细胞形态和存活率变化(噻唑蓝法和Hoechst33324荧光染色法);Western blot检测细胞色素C来反映谷氨酸对神经元的损伤作用,同时观察白黎芦醇对抗损伤的机制。结果谷氨酸对海马CA1区神经元产生强烈的兴奋性毒性,导致神经元大量凋亡和死亡;20μmol·L-1的白黎芦醇对正常神经元的形态和存活率均无明显影响,但能对抗谷氨酸损伤和减轻谷氨酸的兴奋性神经毒性,同时抑制谷氨酸诱发的细胞色素C表达上调。结论白黎芦醇可通过下调海马CA1区神经元细胞色素C的表达水平,降低谷氨酸的兴奋性毒性,对神经元起到保护作用。Objective To investigate the protective effects and its possible mechanism of resveratrol on the cultured hippocampus CA1 neurons suffered from glutanate neurotoxicity in vitro. Methods The 1.5 mmol .L-1 glutamate was added into the culture medium of primary cultured hippocampus CA1 neurons in vitro to induce the glutamate neurotoxicity model, and then 20μmol . L-1 resveratrol was added into the normal and the glutamate-neurotoxicity culture medium respectively to ob- serve the morphologic change and the viability of cells (methyl thiazolyl tetrazolium test and Hoechst33324 fluorescent stai- ning) ;The cytochrome C was detected by western blot to show the injury effect of glutamate on neurons and observe the protec- tive effects of resveratrol. Results Glutamate could produce strong excitotoxicity on CA1 neurons, and induced a large number of neuronal apoptosis and death. 20 μmol . L- 1 resveratrol had no obvious effects on the morphologic change and the viability of neurons. However,it could against the injury induced by glutamate and lighten the excitotoxicity of glutamate. Meanwhile,it could inhibited the up regulation of cytoehrome C induced by glutamate. Conclusion The resveratrol can reduce the excitotoxicity of glutamate by down-regulating the expression level of cytochrome C and play a protection role on neurons.
