摘 要:本文利用蛋白激酶抑制剂PD98059和H2O2荧光探针H2DCF-DA,探索MAP类蛋白激酶和H2O2在调节ABA抑制水稻幼根生长中扮演的角色及其可能的作用关系.结果显示,ABA以浓度依赖性抑制水稻幼根生长,明显诱导H2O2在伸长区积累.外源H2O2处理可有效模拟ABA抑制水稻幼根生长,该结果暗示,H2O2可能位于ABA下游,参与调节水稻幼根生长.蛋白激酶抑制剂PD98059处理,明显缓解ABA诱导的H2O2产生和对水稻幼根生长的抑制;并逆转ABA诱导的过氧化氢酶(CAT)活力上升.这些结果表明MAP类蛋白激酶通过调节根中H2O2的产生介导ABA调节的水稻幼根生长.The possible links among ABA, ROS, and MAP kinase during rice seedling root growth were investigated by using PD98059, a specific inhibitor of MAP kinase; and H2DCF-DA, a H2O2 fluorescence probe. Results showed that ABA can inhibit primary root growth of rice seedlings in dose-dependent manner. It induced H2O2 accumulation in elongation zone, H2O2 mimic ABA inhibiting primary root growth, which indicates that H2O2 may act downstream of ABA signaling in the repression of root growth. PD98059 alleviating ABA-induced H2O2 accumulation, the repressioh of root growth, and activation of CAT. These results suggest that MAP kinase is involved in ABA-regulated primary root growth of rice via modulating H2O2 production in elongation zone and meristem zone.
