摘  要:目的筛选制备最佳补骨脂素中空黏附微球的工艺处方。方法采用Box-behnken实验设计优化、筛选制备最佳补骨脂素中空黏附微球的工艺处方,并应用Design-Expert(8.0.4,试用版)软件分析试验结果。结果所得优化后的处方是投药量为12.36%(Y1)、乙基纤维素/EudragitEPO(EC/Eu)比值为2.27、PVA的浓度为1.53%,制备的补骨脂素中空黏附微球的药物含量为9.18%(Y1),6 h的百分释放率为76.15%(Y2),球形度为80.97%(Y3)。结论所用方法筛选制备补骨脂素中空黏附微球的最佳处方是可行的。OBJECTIVE To optimize the formulation parameters of psoralen loaded hollow - bioadhesive mierospheres. METHODS Box - behnken experimental design was employed to statistically optimize the formulation parameters of psoralen loaded hollow - bioadhesive microspheres. A three- factor, three -level design of experiment with response surface methodology was run to evaluate the main and interaction effect of several independent formulation variables that were included. RESULTS The optimized micro- spheres were obtained with a drug loading( Y )of 9.18%, cumulative drug release in 6 h (Y2) of 76.15% , and spherisity (Y3)of 80. 97% ,respectively. When the drug fed amount ( X1 ) , Ee : Eu ( X2 ) , and PVA ( X3 ) values were 12.36% , 2.27 and 1.53% , respectively. CONCLUSION The results indicated Box -behnken experimental design facilitates the formulation and optimization of psoralen loaded hollow -bioadhesive mierospheres.
