拓扑替康对小细胞肺癌NCI-H446裸鼠移植瘤的药效学研究

摘　　要：目的研究拓扑替康的不同给药方案对荷NCI—H446瘤裸鼠的治疗效果和毒性情况，确定拓扑替康作为阳性药对荷NCI～H446瘤裸鼠的最佳给药方案。方法建立荷NCI—H446瘤裸鼠模型，给予不同剂量和给药间隔的拓扑替康，记录各组裸鼠皮下移植瘤的生长情况、荷瘤鼠的死亡和体重变化情况，计算相对肿瘤体积、肿瘤相对增殖率，绘制肿瘤生长曲线和体重变化曲线。结果与结论每周2次于裸鼠的尾静脉iv5mg·kg-1拓扑替康，间隔2—3d，连续给药3周，拓扑替康对荷NCI—H446瘤裸鼠的治疗效果较好。OBJECTIVE To study the pharmacodynamics and toxicity of Topotecan on small cell lung cancer xenograft model in nude- mouse with different therapeutic regime, in order to establish the best therapeutic scheme of Topotecan as positive control. METHODS The human small cell lung cancer xenograft model was established and the mice were grouped. Then Topotecan was administered with different dosages and different intervals. The tumor growth, animals＇ death and body weight change was recorded. The relative tumor volume and relative tumor proliferation rate were calculated. The tumor growth curve and body weight change curve were plotted. RESULTS and CONCLUSION The regime of 5 mg. kg- l Topotecan tail vein injection twice per week,3 weeks continuously with 2 -3 days＇ interval has the best therapeutic effect, and it is recommended.
