摘  要:目的优化分离获得的一株土壤稀有放线菌发酵培养基,获得该有效成分的大孔树脂分离条件。方法分别利用单因素试验和均匀试验设计法,对发酵培养基构成成分和最佳用量进行优化;分别利用抑菌圈法和液相色谱法对发酵液中有效成分含量进行测定,考察大孔树脂的吸附及解吸附参数。结果经优化后的最佳培养基配方为葡萄糖1.8%、蛋白胨0.5%、豆粕0.3%、酵母膏1.0%、CaCO30.25%,此时发酵液中有效成分活性最强;D-101型大孔吸附树脂对发酵液中有效成分的吸附能力最强,最佳吸附时间为120min,采用70%的丙酮水溶液即可对有效成分形成有效解吸附,最佳解吸附时间为120min。结论通过一次吸附与解吸附分离使发酵液中有效成分得到了有效分离。Objective Optimization the fermentation medium of a soil rare actinomycete sp. and researching the separation condition of the antifungal active ingredient. Methods Based on One-factor experimental design and uniform design method respectively, to optimize the fermentation medium; based on the HPLC and cup-plate method to detect the antibiotic concentration. Results The optimizing results of culture medium by uniform design method was glucose 1.8%, peptone 0.5%, bean cake 0.3%, yeast extract 1.0%, calcium carbonate 0.025%. The dynamic adsorption and desorption study show that D-101 resin had good adsorption ability from fermentation broth, and the optimum adsorption and desorption conditions were as follows: the adsorption and desorption time was 120 minutes, the desorption concentration of acetone was 70%, the pH value of desorption was 6.0. Conclusion The antibiotic production was improved significantly and the antifungal antibiotic was refined efficiently after once adsorption and desorption procedure.
