摘  要:本研究旨在分析TAP1基因在断奶仔猪抗性型和敏感型个体间的表达差异,探讨TAP1基因在断奶仔猪抗F18大肠杆菌中发挥的潜在作用。挑选35日龄苏太猪F18大肠杆菌抗性型和敏感型仔猪各8头,运用实时荧光定量PCR技术对TAP1基因在11个组织中的表达水平进行测定。结果表明:TAP1基因在抗性型和敏感型个体中的组织表达谱呈现一致性,且在肠道组织(十二指肠、空肠)、免疫组织(脾、淋巴结、胸腺)和肺中表达量较高。在11个组织中,抗性型个体的TAP1基因表达量普遍高于敏感型个体,差异倍数从1.21到2.37不等,其中在脾、肺、淋巴结、胸腺和空肠、十二指肠中,抗性型个体的TAP1基因表达量显著高于敏感型个体(P<0.05)。因此,TAP1基因在免疫组织及肠道内的高表达可能对断奶仔猪抵御E.coli F18的感染具有重要作用。This paper aimed at investigating the differential expression of TAP1 gene between Escherichia coli F18- resistant and -sensitive piglets, and exploring the potential role of TAP1 gene in E. coli F18 resistance of weanling pigs. The expression level of TAP1 gene in 11 tissues, which was obtained from eight E. coli F18-resistant and eight E. coil F18- sensitive Sutai piglets of 35 days old, was detected by the method of Real-time PCR. The result showed that the expression profile of TAP1 gene was consistent between sensitive and resistant pigs, and TAP1 expressed much higher in intestinal tissues (duodenum, jejunum), immune tissues (spleen, lymph node, and thymus) and lung. The expression of TAP1 gene in resistant individuals was higher than that in the sensitive ones with the fold change from 1.21 to 2.37. The difference was significant in spleen, lung, lymph node, thymus, jejunum, and duodenum (P〈 0.05). In conclusion, the high expression level of TAP! gene in intestinal tissues and immune tissues played an important role in the resistance to E. coli F18.
