摘 要:目的:观察通过劳倦加饮食不节(高糖高脂膳食)复合腹腔注射链脲佐菌素造模方法能否成功复制出脾气虚证2型糖尿病动物模型。方法:健康雄性Wistar大鼠60只,体重(220±20)g,按体重随机分成二组:空白对照组(n=20)和模型组(n=40)。模型组单日喂食甘蓝15~20g/只,自由饮水,游泳至耐力极限;双日高糖高脂膳食持续喂养,共计12周,第4周后给予链脲佐菌素腹腔注射。结果:模型组大鼠造模开始后,出现了游泳耐力逐日下降,食少懒动,大便溏薄,精神萎靡不振,体毛光亮度减退,从第6周后体重就开始显著增加,链脲佐菌素腹腔注射2周后空腹血糖、餐后2h血糖增高,甘油三酯和总胆固醇分别从第4周后表现出异常,造模结束后模型组出现高血糖症、高胰岛素血症、胰岛素抵抗,符合本研究拟建立的脾气虚证、2型糖尿病的评估标准。结论:通过劳倦加饮食不节(高糖高脂膳食)造模方法可成功复制出脾气虚证2型糖尿病动物模型。Objective : In this study, we observed that whether the modeling approach by fatigue plus improper diet ( high sugar high fat diet) and intraperitoneal injection of streptozotocincan can copy deficiency syndrome T2DM rat model. Methods:60 healthy male Wistar rats,weighing (220 ±20) g,were randomly divided into two groups after adaptive feeding days:blank control group ( n = 20) and model group ( n = 40). The model group were fed cabbage 15x 20 g/rat in a single day and allowed to drink water freely, swimming to the endurance limit. The model group were fed high sugar, high fat diet feeding in a double day, a total of 12 weeks. We looked forward to copy the model of deficiency syndrome T2DM. Results : Rats model showed swimming endurance was decreased, eating less, being lazy, loose and thin stools, spiritual malaise, and the body degree was diminished. The weight be-gan to increase significantly after 6 weeks. Fasting blood glucose was increased after the intraperitoneal injection of streptozotocin. Triglycerides and high density lipoprotein were abnormal after 4 weeks. Rats showed hyperinsulinemia, insulin resistance lining with spleen deficiency syndrome and T2DM evaluation criteria. Conclusion:The modeling approach by fatigue plus improper diet (high sugar high fat diet) and intraperitoneal injection of streptozotocincan can copy deficiency syndrome T2DM rat model.
