摘  要:目的:观察银杏酮酯分散片对急性血瘀型大鼠血液流变学及相关生理指标的影响,为银杏酮酯分散片的临床应用提供药效和安全性参考依据。方法:80只大鼠随机均匀分为空白组、模型组、银杏酮酯分散片低剂量组(25 mg·kg-1)、中剂量组(55 mg·kg-1)、高剂量组(110 mg·kg-1)和金纳多对照组(95 mg·kg-1),采用大鼠反复灌胃方式预给药,每天1次,连续给药1周。随后采用皮下注射大剂量肾上腺素复合冰水浴制备大鼠急性血瘀模型,并以此模型来评价银杏酮酯分散片对全血黏度、血浆黏度、血沉等血液流变学指标的影响。结果:与空白组比较,模型组大鼠表现出全血黏度(200,30,5和1 s-1)、全血还原黏度、全血相对黏度、血沉、红细胞聚集指数均明显增加(P<0.05)的血液流变学异常现象,而银杏酮酯分散片低、中、高组以及金纳多组均可显著降低血液黏度、全血还原黏度、血沉、全血相对黏度以及红细胞聚集指数(P<0.01或0.05)。结论:银杏酮酯分散片具有改善血瘀证大鼠血液流变学异常的作用。Objective: To investigate the effect of ginkgo ketone ester dispersible tablet on blood rleology of rats with acute blood stasis. Methods: Eighty SpragueDawley rats were used. The rats were intragastrically ad ministrated with ginkgo ketone ester dispersible tablet (25, 55 and 110 rag kgX) or ginaton tablet (positive con trol 95 rag. kg1 ) once a day for 7 d. Then, acute blood stasis was induced by subcutaneous injection of large dose adrenaline and icewater bathing. Results: Compared with control, the rats with acute blood stasis exhibited a sig nificant increase in blood rheology indexes ( P 〈 0.05 ) , including whole blood viscosity ( 200,30,5,1 s 1 ) , re duced and relative viscosity of whole blood, erythrocyte sedimentation rate, and erythrocyte aggregation index. Treatment with each dose of ginkgo ketone ester dispersible tablet attenuated the increased indexes in different ex tents(P 〈 0. 01 or 0.05). Conclusion: Ginkgo ketone ester dispersible tablet can attenuate the abnormalities in blood rheology of rats with acute blood stasis, suggesting that it could be applied in preventing thrombogenesis.
