摘 要:为了优化铁皮石斛原球茎在固体培养茎中的增殖生长条件,为铁皮石斛原球茎大量繁殖提供理论依据,以铁皮石斛原球茎为试验材料,研究硝态氮和铵态氮浓度配比、PO43-浓度、糖种类、蛋白胨浓度和ABA浓度对铁皮石斛原球茎增殖生长的影响。结果表明,在固体培养基培养铁皮石斛原球茎的过程中,将硝态氮、铵态氮的浓度配比设置为25/5、将培养基中PO43-的浓度控制为2.1 mmol/L、以白糖作为碳源、添加3 g/L的蛋白胨、加入0.1 mg/L的ABA时有利于铁皮石斛原球茎的增殖生长。In order to optimize the conditions of proliferation of protocorm-like bodies (PLB) of Dendrobium candidum by solid culture, provide theoretical foundation for mass propagation of PLB, protocorms were used as experimental materials to study the effect of NO3-/NH4+, PO43 concentration, sugar type, peptone and ABA concentration on the growth of PLB of Dendrobium candidum. The results showed that the proliferation and growth of PLB were best when the NO3-/NH4+ was 25/5, the concentration of PO43- was 2.1 mmol/L , white sugar was used as carbon source, peptone concentration was 3 g/L and supplemented ABA was 0.1 mg/L during solid culture.
