摘 要:以西南桦种子无菌萌发苗的顶芽和带叶茎段为外植体,探讨TDZ、NAA、琼脂及糖浓度对西南桦组织培养的影响,建立西南桦以芽繁芽的快繁方法。结果表明,以3/4MS+2.5%蔗糖+0.65%琼脂+肌醇0.1 mg/L为芽诱导基本培养基,添加TDZ 0.025 mg/L+NAA 0.015 mg/L的条件下,顶芽外植体增殖效果最好,40 d增殖倍数为4.8;添加TDZ 0.025 mg/L+NAA 0.005 mg/L的条件下,带叶茎段外植体芽增殖效果最好,40 d增殖倍数为2.8,两种外植体增殖系数之和达到7.6。生根试验结果表明,3/4MS+2%蔗糖+0.65%琼脂+肌醇0.1 mg/L为生根基本培养基,添加0.1 mg/L IBA生根效果好。A complete system of rapid-propagation of Betula alnoides Buch Ham. ex D. Don was established, as influences of different concentration of TDZ, NAA, agar, sucrose on their regeneration were investigated. Apical buds and stems with primary leaf, elite of germinated seeds, were used as explants for in vitro rapid propagation by budding. The results showed that 0.65% agar+sucrose 25 g/L+inositol 0.1 rag/L, 3/4MS medium supplemented with TDZ 0.025 mg/L and NAA 0.015 mg/L resulted in best propagation coefficient (4.8 per 40 d) for apical buds; supplemented with TDZ 0.025 mg/ L and NAA 0.05 mg/L resulted in best propagation coefficient (2.8 per 40 d) for stems with primary leaf. Total propagation coefficient was 7.6 per 40 d when two kinds of explants from one seed had been used. With 0.65% agar+sucrose 20 g/L+ inositol 0.1 mg/L, 3/4MS medium supplemented with IBA 0.1 mg/L was suitable for root induction of B. alnoides Buch Ham. ex D. Don.
