摘 要:目的:研究8-羟基二氢小檗碱和小檗碱与多药耐药基因之间的作用。方法:采用MTT比色法测定药物的细胞毒浓度,适时定量PCR测定不同多药耐药基因的表达。结果:在1~100μM浓度范围内,8-羟基二氢小檗碱和小檗碱对Caco-2细胞无毒性作用;8-羟基二氢小檗碱和小檗碱对mdr1和mrp 1的表达作用一致,在25~100μM范围内,小檗碱能明显降低mrp2的mRNA表达,而8-羟基二氢小檗碱对其无明显影响。结论:8-羟基二氢小檗碱与小檗碱对mrp2的基因表达差异较大,其对MRP2蛋白表达的作用以及与基因调控之间的作用关系还有待更进一步的研究。Objective To investigate the effect of 8 - hydroxydihydroberberine and berberine on muhidrug resistance gene using Ca- co - 2 cell model. Method The damage of 8 - hydroxydihydroberberine and berberine to Caeo - 2 cell was evaluated by the MTT meth- od. Expression of muhidrug resistance gene including mdr 1, mrp 1 and mrp 2 was determined by quantitative real time PCR. Result 8 -hydroxydihydroberberine and berberine had no damage to the growth of the Caco -2 cells in the concentrations of 1 ~ 100 μM. 8 - Hydroxydihydroberberine and berberine had the same effects on the expression of mdr 1 and mrp 1 gene. Berberine significantly decreased the expression of mrp 2 gene in the concentration of 25 - 100 μM, but there was no effect in 8 - hydroxydihydroberberine - treated groups. Conclusion The difference on the expression of mrp 2 of 8 - hydroxydihydroberberi - ne and berberine may imply to further study the effects on the proteins and the relationship of them.
