摘  要:目的:探索2型糖尿病大鼠血管内皮细胞内分泌功能的变化,以及有氧运动和膳食控制对2型糖尿病大鼠血管内皮细胞内分泌功能紊乱的干预作用及其可能机制。方法:从62只雄性SD大鼠中随机抽取8只作为正常对照组(C组),饲以标准普通饲料。其余54只喂饲高糖高脂膳食,并采用腹腔注射小剂量STZ建立2型糖尿病(DM)动物模型,分为DM对照组(DMC组)、DM+运动组(DME组)、DM+膳食控制组(DMD组)、DM+运动+膳食控制组(DMED组)4组。采用交互设计研究方案对T2DM大鼠进行60 min无负重游泳运动或/和膳食控制。13周后测定FPG、血浆AGEs、NO含量、主动脉NOS活性等指标。结果:(1)与C组相比,DM组血清NO含量显著降低,血浆ET-1含量和ET-1/NO比值显著升高。长期有氧运动使T2DM大鼠血浆ET-1含量和ET/NO显著降低,NO含量显著上升(P<0.05);膳食控制使T2DM大鼠血浆NO含量无明显升高,但ET-1含量和ET-1/NO显著降低;有氧运动联合膳食控制对降低血浆ET-1和ET-1/NO有显著交互作用,但对升高NO含量无显著交互作用。(2)与C组相比,DMC组FPG和血浆AGEs含量显著升高,同时主动脉ET-1含量显著升高,tNOS和cNOS活性显著降低,iNOS活性有所升高,但无显著性差异。长期有氧运动显著降低T2DM大鼠FPG,显著升高主动脉cNOS活性,但对降低血浆AGEs、主动脉ET-1含量和tNOS、iNOS活性无显著性作用;膳食控制显著降低T2DM大鼠血浆AGEs含量,显著升高tNOS和cNOS活性,但对降低FPG、主动脉ET-1含量和iNOS活性无明显作用;运动联合膳食控制对降低T2DM大鼠FPG、AGEs、主动脉ET-1含量和iNOS活性以及升高tNOS和cNOS活性均无显著交互作用。结论:(1)糖尿病大鼠血浆AGEs含量显著升高,血管内皮细胞损伤而使糖尿病大鼠血管内皮细胞内分泌功能发生紊乱。(2)有氧运动和膳食控制降低T2DM大鼠AGEs生成,减轻内皮细胞损伤,改善糖尿病大鼠血管内皮内分泌功能紊乱。Objective To explore the change in endocrine function of vascular endothelial cells in type 2 diabetic rats after aerobic exercise and diet control. Methods 8 male SD rats were used as normal control group (group C,fed with normal standard diet),and other 54 rats were used to establish type 2 diabetes mellitus model (DM)through intraperitoneal injection of small dose of STZ based on feeding high-carbohydrate and high-fat diet. The rats with DM were divided into following subgroups:DM control group (DMC,n=9), DM+exercise group (DME, n= 10), DM+diet group (DMD, n= 10), and DM+ exercise+diet group (DMED,n=10). 60-minute swimming per day or/and dietary control were conducted to the DM rats. After 13-week experiment,the fasting plasma glucose (FPG),plasma advanced glycation end-products (AGEs), nitric oxide (NO)content, endothelin- 1 (ET-1), and aortic nitric oxide synthase (NOS)activities were measured. Results (1) As compared with group C,the serum NO content decreased,plasma ET-1 content and ET-1/NO increased significantly in group DM. Plasma ET-1 content and ET/NO significantly decreased and NO significantly increased in group DME. There was significant decrease in ET-1 content and ET-1/NO ,but no significant change in serum NO concentration in group DMD. There was significant interaction On reducing the plasma ET-1 and ET-1/NO in group DMED,but no significant interaction on increasing NO content. (2) As compared with group C, FPG, plasma AGEs content,and aortic ET-1 content increased significantly,and tNOS and cNOS activities decreased significantly in group DME,but no significant change in iNOS activity. FPG reduced significantly and aortic cNOS activity increased significantly in group DME ,but no significant changes in plasma AGEs,ET-1 content,and aortic tNOS and iNOS activities. Plasma AGEs content reduced significantly, and aortic tNOS and cNOS activities increased in group DMD ,but no change in FPG,aortic ET-1 content and iNOS activity. There was
