摘 要:目的探讨微电极阵列(microelectrode arrays,MEA)技术检测犬心房电生理特征的价值。方法健康比格犬14只随机分为假手术组和起搏组各7只,假手术组植入起搏器不起搏,起搏组植入起搏器持续起搏左心耳;8周后开胸取出2组心脏,剪取左心耳立即切片(厚度500μm),将标本固定在MEA记录系统,记录2组左心耳组织的场电位波形、场电位时程改变及电激动的传导并进行比较。结果 14只实验犬均成功植入埋藏式动物实验用高频起搏器(400次/min);起搏组连续起搏期间,心电图监测可见起搏器信号,起搏器工作正常,并可见典型心房颤动改变;MEA记录系统显示,2组左心耳场电位形态与倒置心房肌动作电位的膜电位曲线相似;假手术组场电位时程((192.14±15.49)ms)高于起搏组((164.29±7.87)ms)(P<0.01);假手术组电激动传导顺序从左至右、左下至右上向外周缓慢递减传导,传导过程中场电位信号电压逐渐递减;起搏组电激动传导顺序总体趋势与假手术组一致,向外周传导,但在传导过程中电压逐渐降低的梯度样改变消失。结论 MEA技术可用于研究心房电生理特性。Objective To explore value of microelectrode arrays (EMA) technique to the detection of the atrial electrophysiological features in beagle dogs. Methods Fourteen beagle dogs were randomly divided into two groups: pacing group and sham operation group, with 7 dogs in each group. Sham operation group was implanted pacemaker without pacing, and pacing group was implanted pacemaker and was continuously paced the left atrial appendage. After 8 weeks, and the hearts were quickly removed after thoracotomy in two groups. The left atrial appendage was immediately sliced to the thickness of 500 /~m. The samples were fixed in the MEA recording system, then the field potentials, field action potential duration (fAPD) change and electric exciting conduction were recorded and compared between two groups. Results Fourteen dogs were successfully implanted high-frequency pacemakers with buried animal experimental type (400 beats/rain). During continuous pacing, pacemaker worked properly and typical atrial fibrillation changes were found. According to the dispIaying in MEA recording system, field potential morphology of left atrial appendage was similar to the inverted atrial action potential membrane potential curves in two groups, fAPD was higher in sham operation group ((192. 14±15.49) ms) than that in pacing group ((164. 29±7. 87) ms) (P〈0.01). The activation conduction sequence decreased slowly from left to right and from lower left to upper right in sham operation group, and the voltage of field potential signal decreased gradually. The overall trend of electrical impulse conduction to peripheral conduction in pacing group was consistent with sham operation group. But the voltage of gradient decreasing disappeared in the transmission process. Conclusion MEA can be used to study the atrial electrophysiology.
