摘  要:为明确生产用培养基对猪丹毒杆菌G4T10株安全性及免疫原性影响,采用2种不同批次的肉肝胃膜消化汤(20120123批和130226批)及马丁琼脂(20120904批和130129批)进行猪丹毒杆菌G4T10株培养和选菌,然后进行安全及免疫原性试验。结果共筛选出4种不同优势菌落,其中一株的菌落(20120123批肉肝胃膜消化汤+20120904批马丁琼脂)形态最好,具有良好的安全性及免疫原性(小鼠全试验9/10存活,其免疫后攻毒达9/9保护);而有一株菌落(20120123批肉肝胃膜消化汤+130129批马丁琼脂)形态最差,其安全性及免疫原性试验结果差(小鼠安全试验仅6/10存活,其免疫后攻毒仅达2/5保护)。试验表明,不同批次生产用培养基对猪丹毒活疫苗(G4T10株)质量影响大。To determine the effect of the culture media on the safety and immunogenicity of Erysipelothrix rhusiopathiae strain G4Tl0, the strain G4TI0 was cultured and selected in two different batches of meat - liver - gastricmueosa digestion (B20120123 and B130226) and Martin agar (B20120904 and B130129). The safety and immunogenicity of the harvest bacteria were then investigated. Results showed that four kinds of dominant colonies were choosed. One of the colonies from B20120123/B20120904 had the best shape. Its safety was 9/10 survival on inoculation to mice, and the protective efficacy was 9/9 survival post challenge. Another colony from B20120123/B130129 was the worst. The safety was 6/10 survival on inoculation to mice, and the protective efficacy was only 2/5 post challenge. It can be concluded that different batchs of the culture media have large influence on the quality of swine erysipelas live vaccine (strain G4T10).
