摘  要:以 5 -氟尿嘧啶 (5 - Fu)为药芯 ,硅橡胶管为骨架 ,制成 5 - Fu控释植入剂。该控释剂为直径 2 mm、长 2 5 m m的管型胶囊 ,壁厚 0 .5 mm。每只胶囊含 5 - Fu l3.2 mg。用三重蒸馏水作释放介质 ,行体外释药试验 ,高效液相色谱(HPL C)测定 5 - Fu释放量。结果表明控释制剂可保持药物零级释放 ,恒定释药量为 40μg/ d左右 ,时间长达 2 4周以上。The sustained release of anticancer agents from silicone pellets was investigated in vitro.Tube shape pellets containing 13.2 mg of 5 fluorouracil were made of silastic silicone tubes of 2 mm in diameter,0.5 mm in wall thickness and 25 mm in length. Each pellet was immersed in 100 ml distilled water at 37℃.Every 24 hours, the outer fluid was collected and stored at -20℃ until the measurement.The concentrations of 5 fluorouracil were measured by HPLC.Results showed that the controlled release implant maintained a steady zero order release rate about 40 μg/d,lasting 24 weeks.
