葛根素对IL-1β损伤大鼠关节软骨细胞增殖的影响

摘　　要：目的观察葛根素(Pue)对IL-1β损伤大鼠关节软骨细胞增殖的影响。方法取10日龄大鼠膝关节软骨做体外细胞培养,培养后软骨细胞随机分为六组:正常组、IL-1β组、地塞米松组、Pue50组、Pue100组、Pue200组。其中,IL-1β组、地塞米松组、Pue50组、Pue100组、Pue200组分别加入IL-1β(5μg/L)10μmol、IL-1β(5μg/L)10μmol+地塞米松10-9mol/L、IL-1β(5μg/L)10μmol+Pue 50μmol/L、IL-1β(5μg/L)10μmol+Pue 100μmol/L、IL-1β(5μg/L)10μmol+Pue200μmol/L,使用酶标仪测定570 nm光吸收值,计算各组软骨细胞的增殖率。结果正常组、IL-1β组、地塞米松组、Pue50组、Pue100组、Pue200组软骨细胞的增殖率分别为0、-11.044%、0.471%、-2.424%、2.828%、5.387%,地塞米松组、Pue50组、Pue100组、Pue200与IL-1β组比较,差异有统计学意义(P<0.05),且随着Pue剂量升高,使增殖率逐步上升。结论葛根素具有一定的促进软骨细胞增殖的作用,为防治骨性关节炎提供理论依据。Objective To study the influence of Puerarin （Pue） on proliferation of rat chondrocytes injuried by IL-1β.Methods The chondrocytes in vitro got from articular cartilage of 10 days old rat.Cultured chondrocytes were randomly divided into 6 groups,including normal group,IL-1 β group,Dexamethasone group,Pue 50 group,Pue 100 group,Pue 200 group.The IL-1 β group,Dexamethasone group,Pue 50 group,Pue 100 group,Pue 200 group were treated with IL-1β （5μg/L） 10 μmol,IL-1β （5 μg/L） 10 μmol＋Dexamethasone 10-9 mol/L,IL-1β （5μg/L） 10 μmol＋ Pue 50 μmol/L,IL-1β （5μg/L） 10 μmol＋Pue 100 μmol/L,IL-1β （5 μg/L） 10 μmol＋Pue 200 μmol/L.Slected wavelength 570 nm,measured the absorbed value by UV spectrophotometry,and proliferation rate of chondrocytes were calculated.Results The proliferation ratio of chondrocyte of IL-1 β group,Dexamethasone group,Pue 50 group,Pue 100 group,Pue 200 group were 0,-11.044%,0.471%,-2.424%,2.828%,5.387% respectively.The Dexamethasone group,Pue 50 group,Pue 100 group,Pue 200 group were superior to the group of IL-1β （P 〈 0.05） severally.And with the Pue dose increased,the proliferation rate was rising gradually.Conclusion Pue can markedly promote the proliferation of chondrocytes,and it maybe one of the mechanism to achieve the effect of prevention and treatment of osteoarthritis.
