摘 要:目的研究阿那日五味散中挥发油组分的不同提取方法的最佳工艺以及β-环糊精包合挥发油的最佳工艺。方法采用水蒸汽蒸馏法和二氧化碳超临界萃取法提取挥发油;采用饱和水溶液法,运用正交试验,以包合物收得率为评价指标筛选最佳包合工艺。结果阿那日五味散水蒸汽蒸馏法提取挥发油的最佳工艺为加入药材8倍量的水提取4 h;二氧化碳超临界萃取挥发油的最佳工艺为A3B3C3D2,即提取罐的压力为25 MPa,提取罐的温度为45℃,提取2.0 h,分离罐Ⅱ的温度为35℃;水蒸汽蒸馏法提取挥发油的最佳包合工艺为A1B1C3,即:挥发油与β-环糊精比例1∶3,包合温度30℃,水浴搅拌2.0 h;二氧化碳超临界萃取的挥发油的最佳包合工艺条件为A1B2C3D2,即:挥发油与β-环糊精用量比例为1∶3,包合温度为35℃,包合时间为2.0 h,挥发油与无水乙醇比例为1∶1。结论二氧化碳超临界萃取挥发油的量是水蒸汽蒸馏法的2倍多,能够极大地提升提取效率,使用β-环糊精包合挥发油可提高挥发油的稳定性,该方法简便易行,适用于大规模生产。Objective To choose the best technology of different extractions and the best β-cycledextrin inclusion technology of olatile oil.Methods The volatile oil was extracted by steam distillation and supercritical carbon dioxide extraction; the study was carried out with orthogonal design; the inclusion technology was determined by the ratio of inclusion compound.Results The best technology of steam distillation to extract the volatile oil was:adding 8 times amount of water extraction for 3.5 hours; the best technology of supercritical carbon dioxide extraction of volatile oil was A3B3C3D2,that were:25 mPa,45℃,extracted 2.0 hours,the temperature of separate 2 was 35℃; the best inclusion process of the volatile oil extracted by steam distillation was A1B1C3,that were:the ratio of volatile oil and β-CD was 1∶5,the inclusion temperature was 30℃,the inclusion time was 2.0 hours; the best inclusion process of volatile oil extracted by supercritical carbon dioxide extraction was A1B2C3D2,that were:the ratio of volatile oil and β-CD was 1∶3,the inclusion temperature was 35℃,the inclusion time was 2.0 hours,the volatile oil:anhydrous ethanol was 1∶1.Conclusion The volatile oil content extracted by supercritical carbon dioxide extraction is higher than the steam distillation method for more than 2 times,this process can improve the stability of the inclusion,the method is convenient and can be applied to the large-scale production.
