摘  要:通过Box-Benhnken Design(BBD)实验设计原理,优化硫酸香草醛法的显色条件,测定黑骨藤复方中总萜的含量,并通过BBD实验设计,用龙胆苦苷作为对照品,寻找最佳条件.结果表明,显色最佳条件为0.5 mL体积分数3%香草醛冰乙酸溶液,5 mL质量分数62%硫酸溶液,显色温度70℃,显色时间60min,优化条件下拟合方程的预测值与实验结果符合.龙胆苦苷在4.726~94.523 mg/L范围内线性关系良好,其回归方程为:Y=0.008 40X+0.045 54(r=0.999 24).BBD实验设计为该类实验提供显色条件优化的一种新方法,为黑骨藤复方中总萜的测定奠定良好的基础.The objective of this work is optimizing colour reaction condition of vanillin sulfuric acid by the methods of BoxBenhnken Design (BBD) and determinating the content of total terpene in Periploca forrestil Schltr.compound drug,by using the BBD,gentiopcroside as the reference standard,optimizingthe parameters of this reaction.The obtained results show that the best colour reaction condition is 0.5 mL of 3% vanilin/glacial acetic acid solution,5 mL of 62% sulfuric acid solution,reaction temperature of 70 ℃,reaction time of 60min.The forecasting value of the equation fits well with the experimental results under the optimization condition.The calibration curve is linear (r =0.999 24) in the range of 4.726 ~ 94.523 mg/L with a equation of Y =0.008 40X +0.045 54.Conclusion:BBD can supply a new method to optimize the colour reaction condition rapidly,lay a good foundation for the determination of total terpene in Periploca forrestil Schltr.compound drug.
