摘 要:为研究高山红景天提取物抗肺组织细胞纤维化的作用,将高山红景天根茎提取物,以不同的质量浓度加入到体外培养的转化生长因子β1(TGF-β1)诱导的A549细胞中,观察细胞的形态变化.采用MTT法测定其对细胞增殖的影响,Western Blotting法测定细胞中间充质标志物Fibronectin EDA(Fn-EDA)的表达变化.结果表明:高山红景天提取物使TGF-β1诱导的A549细胞形态向间充质细胞转化,降低了TGF-β1作用的A549细胞中Fn-EDA的表达,促进了TGF-β1诱导的A549细胞的增殖,说明高山红景天提取物对体外诱导的肺组织细胞纤维化状态有明显的改善作用.This experiment was designed to investigate the role of the extracts from Rhodiold sachalinensis A. Bor on the functions and influences against pulmonary fibrosis. The chemical constituents extracted from the Rhodiold sachalinensis A. Bor's rhizome was made up into several groups with different concentrations, and then added to the A549 cells induced by the transforming growth factor beta-1 (TGF-β1). The changes of A549 cellular morphology were observed. The method of Western Blotting was used to identify the expression of the sign of cell mesenchyme-Fibronectin EDA(Fn-EDA),and MTT method was used to determine the cell proliferation. The results showed that the extracts of Rhodiold sachalinensis A. Bor made the A549 cells induced by TGF-β1 inhibit their cellular morphology changing to mesenchyme cell,at the same time,the Fibronectin EDA(Fn-EDA) possessed down-regulated expression,and the A549 cells induced by TGF-β1 proliferation increased. It was testified that the extracts of Rhodiold sachalinensis A. Bor obviously improved the fibrillation of the pneumonocyte induced in vitro. The study was aim at providing the theory on how to screening the effective constituent in Rhodiold sachalinensis A. Bor aginst pulmonary fibrosis.
