摘  要:本研究以解析日本东部地区立克次氏体菌群的生物形态以及基因特征为目的,对该地区立克次体菌种,如日本立克次体、嗜吞噬细胞无形体、查菲埃立克体、米库尔新埃希体暂定种、牛边缘无浆体以及中央无浆体等的宿主野生鹿以及媒介蜱虫体内组织提取物进行抗体分析和基因解读。结果从105头野生鹿中检测到了嗜吞噬细胞无形体p44外膜蛋白表达基因的阳性率为95.7%,野鹿型16S rDNA的阳性率为96.2%,牛边缘无浆体和中央无浆体的阳性率分别为58.1%和79.0%。野生鹿中没有测出日本立克次体、查菲埃立克体以及米库尔新埃希体暂定种的目标特征基因。在抗体检测中立克次体的阳性率为73.9%,嗜吞噬细胞无形体的阳性率为25.0%。对T-162,T-176蜱虫唾液腺DNA提取物的分析结果证明了在媒介蜱虫体内嗜吞噬细胞无形体,米库尔新埃希体暂定种及中央无浆体的混合感染。这也是日本首次从蜱虫体内测出米库尔新埃希体暂定种。这些发现给立克次体感染症的预防以及深入研究提供了重要的科学参考依据。To investigate the natural morpho!ogy and genetic characters of the rickettsiae pathogen from east area of Japan, antibody analysis and genetic analysis for R. japonica, A. phagocytophilum, E. chaffeensis, Candidatus N, mikurensis, A. bovis and A. centrale were performed on the extracts of the deer spleen and Ticks body, the suspicious carriers isolated from east Japan. A total of 105 deer were investigated and 95.7% of them were found to be positive for A. phagocytophilum p44 outer membrane protein encoding gene. A- mong them, positive rate for deer type 16S rDNA was 96.2% , tile positive rate for A. bovis and A. centrale was 58.1% and 79.0%. The gene of R. japonica,E, chaffeensis and Candidatus N. mikurensis were not detected from deer. For the antibody analysis, the posi- tive rate for Rickettsia and A. phagocytop|filum was 73.9%, and 25.0%, respectively. By investigating salivary gland DNA extraction of Ticks T- 162 and T- 176, multiple infections of A. phagocytophilum,Candidatus N. mikurensis and A. centrale were found in Ticks sample. This is the first study report the infection of Candidatus N. mikurensis in Ticks from Japan. The finding of this study could form a base for the prevention of rickettsiasis in Japan.
