摘 要:日的研究消渴丸对大鼠2型糖尿病的治疗作用,并与格列本脲进行对照研究。方法100只雄性SD大鼠,取12只为正常对照组,其余大鼠给予高脂饲料。4周后以链脲佐茵素(STZ)30mg/kgip,将随机血糖≥11.1mmol/L并剔除血糖过高或胰岛素过低的大鼠,按血糖(BG)水平随机分为:模型组,消渴丸高、低剂量组(1、0.5g/kg·d),格列本脲高、低剂量组(1、0.5mg/kg·d),每组12~13只,灌胃给药,连续12w。观察药物对糖尿病大鼠体重、尿量、摄食量、饮水量、BG、糖化血红蛋白(HbA1c)、胰岛素(Ins)、胰岛素抵抗指数(HOMA—IR)的影响。结亲与正常组相比,模型组大鼠BG、HbA1c、HOMA—IR、尿量、摄食量、饮水量显著升高,Ins、体重显著下降。与模型组相比。消渴丸高、低剂量组组和格列本脲高、低剂量组大鼠BG、HbA1c、HOMA—IR、尿量、摄食量、饮水量明显下降,Ins和体重明显升高。与格列本脲组高、低剂量组比较,消渴丸高、低剂量组组大鼠BG、HOMA—IR和HbA1c进一步降低(P〈0.05)。结论消渴丸(1、0.5s/kg·d)灌胃给药对大鼠2型糖尿病有良好的治疗作用,消渴丸的降糖和改善胰岛素抵抗作用优于等量的格列本脲。Objective to study the therapeutic effect of Xiaoke pill on rats with type 2 diabetes mellitus, and compared with glibenclamide. Methods 100 male SD rats, 12 rats as the normal control group, the other rats were fed with high fat diet. After 4 weeks in streptozotocin (STZ) 30 mg/kg IP, the random blood sugar ≥ 11.1 mmol/L and removed the high blood glucose or insulin low rat, according to blood glucose (BG) level were randomly divided into three groups: model group, high, low dose group of Xiaoke pill ( 1,0.5 g/kg ~ d), glibenclamide, low dose group ( 1,0.5 mg/kg, d), each group of 12 ~ 13 rats, intragastric administration, for 12 W. Observed the effect of drugs on diabetic rat body weight, urine volume, food intake, water intake, BG, glycosylated hemoglobin (HbAlc), insulin (Ins), insulin resistance index (HOMA -IR) effect. Results compared with the normal group, model group rats BG, HbA1c, HOMA -IR, urine volume, food intake, water intake was significantly increased, Ins, body weight decreased significantly. Compared with the model group, high, low dose group of Xiaoke pill group and glibenclamide high, low dose group rats BG, HbA1 c, HOMA -IR, urine volume, food intake, water intake and body weight decreased significantly, Ins increased sig- nificantly. Compared with glyburide group, low dose group, low dose group of Xiaoke pill high, BG, HOMA - IR and HbA1 c group rats were further reduced ( P 〈 0.05). Conclusion Xiaoke pill (1,0.5 g/kg·d) by intragastric administration to the rats with type 2 diabetes mellitus has a good therapeutic effect of Jiangtang Xiaoke pill, and improve insulin resistance effect is better than an equal amount of glibenclamide.
