摘 要:为研制H3N2亚型犬流感病毒灭活疫苗,本实验通过鸡胚传代的方法培育出一株高繁殖能力的鸡胚适应株CGD1株。分别应用白油和氢氧化铝胶佐剂制备两种灭活疫苗,通过监测疫苗免疫后BALB/c小鼠和比格犬的抗体水平以及比格犬的攻毒保护试验,评价两种疫苗的免疫效果。结果表明,两种佐剂疫苗均能够诱导小鼠和犬产生较强的免疫应答反应,二免后14 d,免疫犬抗体水平均高于1∶40;攻毒试验中,疫苗免疫可以显著减轻病毒感染犬临床症状,明显缩短病毒的排毒期,特异性抗体水平显著提高。本研究研制了两种佐剂H3N2亚型犬流感灭活疫苗,为犬流感的防制提供保障。To evaluate the immunogenicity and protective efficacy of the inactivated H3N2 subtype canine influenza virus (CIV) vaccine emulsified in mineral-oil or aluminum hydroxide adjuvant, the BALB/c mice and Beagle pups were immunized with the vaccines, respectively, and the pups was challenged with avian-origin H3N2 subtype virulent CIV post vaccination (PI). The results showed that the vaccinated mice and pups developed high titers of hemagglutination inhibition antibody PI. In addition, the vaccinated pups did not show any clinical signs and the duration of virus shedding was significantly shorter than that in the control group after challenge. These results indicated that both the two adjuvant vaccines were able to induce strong immune response and provide protection against the H3N2 subtype CIV challenge in dogs. However, the mineral-oil adjuvant vaccine was superior to the aluminum hydroxide adjuvant vaccine.
