摘  要:为增强肌肉注射免疫条件下猪肺炎支原体(Mhp)活疫苗的免疫刺激能力,并补充其缺乏的针对重要黏附因子P97R1的应答,本实验采用透析法制备含有P97R1的免疫刺激复合物(ISCOM-P97R1),并将ISCOMP97R1作为活疫苗的佐剂,通过小鼠模型进行免疫学评价。结果表明,电镜下观察所制备的ISCOM-P97R1具有ISCOM的典型蜂窝状结构,直径为30 nm^40 nm,P97R1蛋白的包封率可达85.6%。免疫后7 d即可在小鼠血清中检测到高水平的特异性P97R1抗体,并且其针对Mhp全菌蛋白的抗体也显著高于无佐剂的活疫苗对照组。结果表明,ISCOM-P97R1对Mhp活疫苗免疫刺激能力具有显著的增强作用,同时可以有效诱导产生针对P97R1的免疫应答,进一步增强了疫苗的保护能力。To investigate the enhancement of immunostimulating complex containing P97R1 antigen (ISCOM-P97R1) to the immunostimulating effect of the Mycoplasma hyopneumoniae live vaccine via intramuscular injection, as well as its effect to supplement the immune response to the important adhesions P97R1, ISCOM-P97R1 was prepared by dialysis method, which was subsequently used as an adjuvant for the live vaccine. The immunologic effect was evaluated in the mice model. The prepared ISCOM-P97R1, with a size of 30 nm to 40 nm, showed a cage-like structure under electron microscope. The entrapment rate of protein prepared was 85.6%. In addition, One week after the inoculation, the high levels of antibody against P97R1 and whole M.hyopneumoniae proteins could be detected in the serum of the mice immunized with the ISCOM-P97R1 adjuvanted live vaccine, which was significantly higher than that in the mouse group immunized with the live vaccine without adjuvant. In conclusion, ISCOM-P97R1 showed a significant enhancement to stimulate immune response on the live vaccine, and induced the immune response to P97R1 effectively which further enhanced the protection ability of the vaccine.
