摘  要:为研究猫源蓝氏贾第虫EF1α和GDH基因的分子特性,本研究采用两组引物EF1-F、EF1-R和GDHeF、GDHiF、GDHiR通过普通PCR及套式PCR方法对两基因片段进行特异性扩增,用MegAlign软件对扩增片段进行同源性比对及进化树分析。结果显示,获得了200 bp及432 bp的EF1α和GDH序列,其基因登录号分别为KC510661和KC510662,EF1α基因位点分析表明该蓝氏贾第虫分离株为基因型F,GDH基因位点分析表明其可能是一个新亚型(FIII)。该研究首次对猫源蓝氏贾第虫EF1α和GHD基因双位点进行分子特性研究,为贾第虫分子分类学及分子遗传学研究奠定了基础。To study the characteristics of the EFla and the GDH genes of Giardia lamblia isolated from cat, the two gene fragments were amplified using normal and nested PCR with the primers EFI-F/EF1-R and GDHeF/GDHiF/GDHiR designed according to Giardia sequence in GenBank. The PCR amplicons were sequenced and analyzed by multiple sequence alignment with MegAlign. The results showed that the length of EFlo~ and GDH gene were 200 bp (KC510661) and 432 bp (KC510662), respectively. The EFlc~ gene analysis indicated that the cat-derived G.lamblia belonged to genotype F, and the GDH gene analysis showed that it might be a new sub-genotype (FIII), which provided a foundation on molecular identification and molecular genetics of the G.lamblia in China.
