摘  要:目的:探索沉默NAALADase基因的方法对骨髓间充质干细胞(BMSC)静脉移植,评价其对大鼠脑损伤后的神经保护作用。方法:体外分离培养BMSC,应用小分子干扰RNA转染方法沉默NAALADase基因的表达。建立脑损伤模型后,48 h处死动物取脑组织进行Fluom-Jade B组织荧光染色以及HE染色。结果:HE染色显示沉默NAALADase基因BDNF数目明显增多并且损伤后神经元退变也减少。结论:沉默NAAG肽酶的表达后能有效实现对神经的保护作用。Objective: To determine whether the NAALADase gene silencing can enhance curative effects of intravenous trans- plantation of bone marrow derived stroma cells (BMSCs) on traumatic brain injury (TBI) in rats. Methods: BMSCs were cultured by the suspension culture in vitro, and were transfected by siRNA to knock down the expression of NAALADase. The models of severe TBI were made in rats by moderate lateral fluid percussion. Rats were euthanized at 48h after TBI and brains were processed and stained ac- cording to the Fluom-Jade B Histofluorescence and HE solution. Results: HE histochemical staining results showed that the density of BDNF in the NAALADase gene silencing group was significantly higher and the group reduced the ipsilateral astrocyte loss. Conclusion: The BMSCs ofNAALADase gene silencing transplanted by vein into the traumatic brain tissues can significantly improve the neurologi- cal function in rats with TBI.
