摘  要:目的观察还脑益聪方对快速老化小鼠(SAMP8)学习记忆能力、脑组织海马病理形态及脑组织氧化应激和炎性因子含量的影响。方法采用雄性快速老化小鼠为实验动物,随机分为5组:模型组、阳性药物组(盐酸多奈哌齐0.65 mg/kg)和还脑益聪方大、中、小(21.84、10.92、5.46 g/kg)剂量组,每组10只,连续灌胃给予相应药物。采用Morris水迷宫实验观察小鼠行为学的变化。麻醉处死小鼠后,观察小鼠海马病理形态,比色法测定小鼠海马和皮层过氧化物歧化酶(SOD)、丙二醛(MDA)、谷胱甘肽(GSH)、谷胱甘肽过氧化物酶(GSH-PX)的含量及活性,放射免疫法检测小鼠海马及皮层组织白细胞介素-1(IL-1)和白细胞介素-6(IL-6)。结果与模型组比较,还脑益聪方可以增加SAMP8小鼠穿台次数,延长小鼠在第四象限的路程和时间(P<0.05);改善SAMP8小鼠脑组织病理形态,增加小鼠海马和皮层中SOD和GSH-PX的活性(P<0.05,P<0.01),降低IL-1、IL-6水平(P<0.05,P<0.01)。结论还脑益聪方通过抑制SAMP8小鼠脑内氧化应激与炎症反应,保护脑组织海马和皮层的结构形态,从而提高SAMP8小鼠空间识别和学习记忆能力。Objective To observe the influences of Huannao Yicong Fang on learning and memory ability, hippocampal pathomorphology and content of cerebral oxidative stress and inflammatory factors in rapid aging SAMP8 mice. Methods group, positive drug group ( donepezil dose (10.92 g/kg) and low-dose (5. dose group and low-dose group, each medicinal continuously. The changes Male rapid aging SAMP8 mice were randomly divided into model hydrochloride, 0.65mg/kg) and high-dose (21.84 g/kg), mid- 46 g/kg) Huannao Yicong Fang groups (high-dose group, mid- n = 10), and all groups were intragastrically given corresponding of mouse ethology were observed by applying Morris water maze. The content and activity of hippocampal and cortex superoxide dismutase (SOD), malondialdehyde (MDA), glutathione and glutathione peroxidease (GSH-PX) were detected by using chromatoptometry. The levels of interleukin-1 ( IL-1 ) and interleukin-6 ( IL-6 ) were detected by using radioimnmnoassay. Results Compared with model group, Huannao Yicong Fang increased the number of passing platform,prolonged the distance and time in the fourth quadrant (P 〈 0. 05 ), ameliorated the cerebral pathomorphology, increased the activity of SOD and GSH-PX (P 〈0. 05, P 〈0.01 ), and decreased the levels of IL-1 and IL-6 ( P 〈 0. 05, P 〈 0.01 ). Conclusion Huannao Yicong Fang can improve the abilities of space recognition, learning and memory through inhibiting oxidative stress and inflammatory reactions and protecting the morphology of cerebral hippocampus and cortex in rapid aging SAMP8 mice.
