摘 要:采用ISSR分子标记技术对来自广东、广西及海南3省的19个居群57份美丽鸡血藤种质及其易混淆的3份近似种种质的遗传多样性和亲缘关系进行研究,旨在为美丽鸡血藤种质资源的鉴定、保护和开发利用提供参考.结果表明:8条ISSR引物在美丽鸡血藤基因组中扩增条带共91条,其中多样性条带76条,占84.4%;Nei,s基因多样性指数(H)和Shannon信息指数(Ⅰ)分别为0.258 3和0.396 0,这表明美丽鸡血藤种质的多样性丰富;群体遗传分化系数(Gst)和基因流(Nm)分别为0.651 5和0.267 5,这表明居群间遗传分化明显,基因流水平偏低:聚类分析结果显示所有美丽鸡血藤种质主要分为2大类,亲缘关系与地理分布并不具有明显的一致性.4个近似种间的遗传距离为0.235 2~0.416 2,其中以广东鸡血藤与美丽鸡血藤的亲缘关系最近,筛选出的4条ISSR引物能对这4个物种进行有效区分,可用于物种的鉴别分析.To study the genetic variation and genetic relationship of Callerya speciosa and provide reference information on the identification,conservation and utilization of the germplasm resources,fifty-seven individuals of 19 population of C.speciosa collected from Hainan,Guangdong and Guangxi,as well as its three relative species,were analyzed by ISSR markers.A total of 91 bands were scored from the 8 primers,and 76 of them were polymorphic,which accounted for 84.4%.At the species level,the Nei's genetic diversity(H) and the Shannon information index(Ⅰ) was 0.258 3 and 0.396 0,respectively,reflecting the high diversity within the accessions of C.speciosa.The coefficient of genetic differentiation among population(Gst) and gene flow was 0.651 5 and 0.267 5,indicating strong genetic differentiation and low gene flow among the population.Cluster analysis revealed that all the samples of C.speciosa could be divided into two major classes but not corresponding to the biogeographical distribution of the species.The genetic distance among the four relative species ranged from 0.235 2 to 0.416 2 and C.fordii showed the closest genetic relationships with C.speciosa than other two species.Four ISSR markers selected in the study could be used as a rapid molecular diagnostic technique for species identification because of the obvious different amplified bands in these four species.
