摘　　要：利用改良的SDS-醋酸钾沉淀法可以有效分离纯化胶乳DNA。胶乳经过高速离心后分为3层,上层主要是橡胶粒子,中间层是胶乳的可溶性胞质成分即C-乳清,下层主要是黄色体。分别对这3层组份进行DNA提取分析,结果显示胶乳DNA主要存在于黄色体层。限制性内切酶分析表明所得胶乳DNA是序列不均一的DNA分子。胶乳DNA主要由线粒体DNA和少量细胞核DNA组成。健康树中胶乳DNA含量要大于死皮树胶乳DNA含量。The total latex DNA in Hevea brasiliensis could be obtained by the improved SDS-KAc precipitating method.After high centrifugation the Hevea latex was separated into three main fractions：the rubber cream at the top,the aqueous C-serum and the bottom fraction-lutoids.We extracted and analyzed the DNA concent of the rubber cream,the aqueous C-serum and the lutoids fraction.Results showed that the latex DNA mainly existed in the lutoids fraction.Restrict endonucleases analysis of latex DNA demonstrated that latex DNA were heterogeneous DNA Sequences.The analysis of PCR results indicated that the latex DNA probably contained mainly mitochondrial DNA and some nucleus DNA.In addition,the latex DNA content from the healthy rubber trees was higher than that from the TPD trees.
