摘  要:目的建立酒精诱导神经元凋亡的体外模型,探讨酒精性痴呆的发病机制。方法以SH-SY5Y细胞为实验对象,通过维甲酸(RA)诱导细胞分化,之后以不同浓度的酒精作用于细胞,连续培养24h。四甲基偶氮唑蓝(MTT)比色法检测细胞增殖;碘化丙啶(PI)染色观察细胞凋亡。结果 RA诱导分化后SH-SY5Y细胞表现神经元形态特征,酒精抑制SH-SY5Y细胞增殖并呈浓度依赖关系(P<0.001)。酒精(100mol/L)作用24h后明显诱导细胞凋亡。结论分化后SH-SY5Y细胞对酒精诱导损伤敏感,可用于建立酒精性痴呆体外研究模型。Objective To study the effect of ethanol on induction of apoptosis in differentiated SH-S^5Y cells. Methods SH-SY5Y cells were induced to differentiation with administration of retinoie acid (RA) for 6 days. Then cells were tt:eated with different doses of ethanol for 24h. The growth and proliferation of SH-SY5Y cells were analyzed with MT]" assay. The staining with pidium iodide was used to observe the cell apoptosis. Results The cell proliferation was inhibited by ethanol in the dose-dependent manner. Inceased apoptosis cells were detected when cells were treated with 100mol/L concentration of ethanol. Conclusions Ethanol administration may contribute to the induction of apoptosis of SH-SY5Y cells. The model that ethanol induced-SH-SY5Y cell apoptosis may be used for the research of the mechanism of alcohol-as- sociated dementia.
