热毒宁注射液的解热抗炎作用及其机制

摘　　要：目的:探讨热毒宁注射液抗炎、清热解毒的作用及其机制。方法:分别以二甲苯致小鼠耳肿、角叉菜胶致大鼠足趾肿胀、冰醋酸致小鼠毛细血管通透性增加等炎症模型;以内毒素感染家兔致内毒素性发热模型,观测小鼠耳肿胀及大鼠足趾关节肿胀度,检测白介素-1(IL-1)、白介素(IL-6)、内皮素(ET-1)、前列腺素E2(PGE2)等炎症介质。结果:热毒宁注射液10.15 g·kg-1、5.08 g·kg-1剂量组对角叉菜胶致大鼠足趾肿胀均有抑制作用,1 h开始显效,持续至4 h;2.54 g·kg-1剂量组在2 h方显效,抑制作用可持续至4 h;10.15 g·kg-1剂量组对二甲苯致小鼠耳肿胀有明显的抑制作用,并能降低小鼠毛细血管通透性;注射内毒素2 h,模型组家兔肛温值居高不下,但热毒宁2.54 g·kg-1、5.08 g·kg-1两剂量组具有明显清热、降体温的作用,对家兔血清IL-1、IL-6、ET-1、PGE2;大鼠血清IL-1、IL-6等炎症介质均有不同程度的抑制作用。结论:热毒宁注射液对内毒素引起的家兔发热具有解热降温作用;对角叉菜胶致大鼠炎症有明显抑制作用、能显著降低大鼠IL-1、IL-6水平;并能抑制二甲苯致小鼠耳肿胀,提示热毒宁注射液具有较强的抗炎解热作用,其机制可能与抑制IL-1、ET-1、IL-6、PGE2等炎症介质有关。OBJECTIVE To study the anti inflammation and clearing heat and detoxicating effect of Reduning injection and its mechanism of actions. METHODS Inflammation models were established by xylene-induced mouse ear swelling, carrag evnin induced rat paw swelling and glacial acetic acid induced increase of blood capillary permeability in mice, respectively. En dotoxin heating model was induced by infecting the rabbit wilh toxin. The swelling degree of mice ear and rat digits joint was observed. The inflammatory mediums of IL-1, IL-6, endothelin （ET-1）, prostaglandin E2 （PGE2） were detected. RESUUIS Reduning injection with doses of both 10,15 g.kg^-1 and 5.08 g. kg-1 inhibited the rat paw swelling caused by carrageenin, with the effect appearing 1 hour and continuing up to 4 hours after receiving the injection. The inhibitory effect of Reduning injection with a dose of 2.54g·kg-1 dose appeared powerfully in 2 h and sustained to 4 h. Reduning injection with a dose of 10. 15 g·kg-1 had an obvious inhibitory effect on mouse ear swelling induced by xylene, and could reduce capillary permeability of mice. The anal temperature of the rabbits was continuingly high after receiving injection of endotoxin for two hs. Redunmg injection whh doses of both 5.08g·kg-1 and 2. 54g·kg-1 could relieve heat, lower body temperature obviously and inhibit rabbit serum inflammatory inedialors of IL-1, IL-6,endothelin （ET 1）, prostaglandin Ee （PGE2） with different degrees. CONCLUSION Reduning injection had antipyretic and hypothermia effect on fever of rabbit caused by endotoxin, inhibited rat inflammation in- duced by carrageenin obviously, reduced the levels of IL-1 and IL-6 in rat significantly and inhibited mouse auricular swelling induced by xylene. The results indicated that Reduning injection had strong anti-inflammatory and antipyretic effects, its mechanism might correlate with the inhibition of inflammation medium such as IL-1, ET-1, IL-6 and PGE2.
