摘  要:目的:观察云芝糖肽(PSP)对环磷酰胺(Cy)所致免疫抑制小鼠Th1/Th2平衡的影响。方法:18只雄性Balb/c小鼠随机分为3组,每组6只。PSP组予以125 mg·kg-1·d-1的云芝糖肽溶液灌胃,正常对照组及模型组予以生理盐水灌胃作为对照,连续25 d。用药第17天和第21天除正常对照组外均予以腹腔注射环磷酰胺制备小鼠免疫抑制模型,观察小鼠脾组织IL-2、IL-4、T-bet、GATA-3 mRNA相对表达变化。结果:各组小鼠脾组织IL-2、T-bet mRNA相对表达水平之间无明显差异(P>0.05);模型组小鼠IL-4、GATA-3 mRNA相对表达水平明显高于正常对照组及PSP组(P<0.05);正常对照组与PSP组的IL-2/IL-4及T-bet/GATA-3水平相比无统计学差异(P>0.05),但均明显高于模型组(P<0.01)。结论:云芝糖肽可以纠正免疫抑制机体IL-2/IL-4、T-bet/GATA-3的偏移状态,促进机体Th1/Th2的平衡稳定状态,发挥免疫保护作用。OBJECTIVE To investigate the influences of polysaccharidepeptide (PSP) of Coriolus wersicolor on Th1/Th2 balance in the cyclophosphamide-induced immunosuppressive mice. METHODS Eighteen Balb/c mice were randomly divided into 3 groups. The mice in normal control group and model group were orally administrated with saline water, while the PSP group received PSP( 125 mg.kg -1.d-1) treatment for 25 days. On the 17th day and 21st day, all groups except normal control group were injected intrapcritoneally with cyclophosphamide(Cy) (150 mg.kg-1 . d-1) to induce the model. The relative mR NA expressions of mterleukin-2(IL-2), interleukin-4(IL-4), transcription factor T-box-containing protein(T-bet), and GATA binding protein 3(GATA-3) in the spleen tissues of each mice were detected by quantitative real time PCR. RESULTS There were no obvious differences in relative expressions of IL-2 mRNA and T-bet mRNA in each group. However, the IL-4 and GA TA-3 mRNA relative expressions in the model group were significantly higher than the normal control group and PSP group (P 〈0. 05). There were no statistical differences between normal control group and PSP group in the ratios of IL-2/IL-4 and T- bet/GATA-3(P〉0.05), but they were significantly higher than the model group (P〈0.01). CONCLUSION PSP can correct the shifted status of IL-2/IL-4 and T bet/GATA-3 in immunosuppressive mice, improve the balance of Th1/Th2, and produce immuno-protective effect.
