摘  要:目的:观察唐草片对注射氢化可的松所致免疫抑制小鼠免疫功能的影响。方法:BALB/C小鼠60只,雄性,将其随机分6组(正常对照组、模型对照组、阳性对照组、唐草片高、中、低剂量组)。各组连续3周灌服相应的药物,给药2周后,除正常组外,各组连续7 d均腹腔注射氢化可的松造模,BALB/C小鼠腹腔注射氢化可的松,造成免疫功能抑制的动物模型,观察连续灌服唐草片的治疗组与模型组动物免疫功能的差异。检测指标有:外周血细胞计数;胸腺指数、脾脏指数;紫外分光光度法测定碳廓清指数;酶联免疫吸附实验(ELISA)法检测鼠血清中IL-2、INF-γ;免疫组化测定空肠IgA、CD4+和CD8+T细胞的表达量。结果:唐草片中剂量组与模型组相比,能够提高免疫抑制小鼠外周血白细胞(WBC)和淋巴细胞计数(LYM)、胸腺指数、脾脏指数、碳廓清指数、血清中IL-2、INF-γ水平,增加肠粘膜中IgA、CD4+和CD8+T细胞的表达量,均有统计学意义(P<0.05)。结论:唐草片能够提高免疫抑制小鼠机体的免疫功能,对免疫失衡机体具有保护作用。OBJECTIVE To evaluate the effects of Tangcaopian on the immunological functions of immunosuppressive mice. METHODS Sixty female BALB/C mice were randomly divided imo six groups (n = 10): normal, model, positive control, Tangcaopian groups (including low, medium, high dosage). Drugs were given ig daily for 3 weeks. After the completion of 2 weeks of lbe treatment, all the animals except the normal group were given hydrocortisone intraperitoneally for 7 days. 6 hours later of the last treatment, peripheral blood leukocytes and lymphocyte numbers were counted. The index of carbon clearance was tested by ultraviolet spectrophotometry. The expressions of IgA, CD4+ and CD8 + T cells of intestinal mucosa were deter mined by immunohistochemistry. Serum levels of 1L-2 and IFN 7 were quantified by enzyme linked immunosorbent assay. RESULTS Tangcaopian groups especially the medium dosage increased peripheral blood lymphocytes and leukocyte numbers, the index of carbon clearance, thymus and spleen index, proportion of IgA, CD4+ and CD8+ T cells in intestinal mucosa and IL-2, IFN-γ in serum. CONCLUSION Tangcaopian can improve immunological functions of immunosuppressive mice.
